RP-HPLC AND SPECTROPHOTOMETRIC METHODS FOR THE SIMULTANEOUS ESTIMATION OF BUPROPION HCl AND NALTREXONE HCl
AbstractA simple, sensitive, precise, accurate and cost effective reverse phase high performance liquid chromatographic method for the simultaneous estimation of bupropion HCl and naltrexone HCl from a single formulation was developed and validated in accordance to ICH guidelines. The two drugs were eluted under isocratic mode using a 250 x 4.6 mm i.d, 0.45 µm Phenomenex Chromosil C18 column. The mobile phase was composed of a mixture of methanol, acetonitrile and water in the ratio of 60:20:20 v/v (pH adjusted to 4.8) at a flow rate of 1.0mL/min. The limit of detection and limit of quantification were found to be 0.5μg/mL and 1.7μg/mL, for both the drugs. The label claim of the combined dosage form was in good agreement with the experimental assay amounts; 99.5% and 99.7% for bupropion HCl and naltrexone HCl, respectively. This method can be successfully applied for the simultaneous estimation of bupropion HCl and naltrexone HCl combination for routine quality control analysis
