A simple, accurate & precise method was developed for the simultaneous estimation of Darunavir (DAR), Cobicistat (COB), Emtricitabine (EMT), and Tenofovir alafenamide (TEN) in their bulk and pharmaceutical formulation. The chromatogram was run through Agilent C18 (150 × 4.6 mm, 5 m) column. Mobile phase containing 0.1% Ortho Phosphoric acid (OPA) (pH 2.4) and Acetonitrile in the ratio of 55:45 was pumped through the column at a flow rate of 0.75 ml/min. While the temperature was maintained at 30°C. The optimized wavelength for the combination was 245 nm. The retention time of DAR, COB, EMT, and TEN were found to be 3.988 min, 3.147 min, 2.205 min, and 2.616 min respectively. The linearity range for the method was found to be 200-1200µg/ml for DAR, 37.5-225 µg/ml for COB, 50-300 µg/ml for EMT and 2.5-15 µg/ml for TEN with regression coefficients of 0.9992, 0.9997, 0.9997 and 0.9994. The potential of transferosomal formulation for transdermal delivery of Ginger constituents was investigated. The aim was to reduce the dosage and improve patient acceptance. Transferosomes were formulated by ethanol injection technique using Phospholipids (Lipoid P-100), cholesterol, various edge activators (tween 80, Span 80), and other excipients. Transdermal patches were prepared by solvent casting method using polymers (Ethylcellulose, HPMC), penetration enhancers (Oleic acid, Tween 80) and PEG 400 as a plasticizer. Transferosomes were evaluated for in-vitro release, particle size, zeta potential, entrapment efficiency, and stability. Patches were evaluated for folding endurance, uniformity of weight, drug content, and thickness. Transferosomes with 1:2:0.25 ratio of Phospholipid: ginger extract: Tween 80 showed particles of 318.8nm, the zeta potential of -6.3mV and 74.4% entrapment efficiency and 67% in-vitro release. Aframomum melegueta (Alligator pepper) is a tropical plant that belongs to the Zingiberaceae (Ginger) family, consumed for its culinary and health benefits. This study investigated the therapeutic efficacy of ethanolic seed extract of Aframomum melegueta at ameliorating paraquat-induced ovarian toxicity in rat models. Thirty female Wistar rats were divided into control and five test groups (n=5). A simple reverse phase SI-HPLC method was developed for the simultaneous estimation of antidiabetic combination of metformin and gliclazide. The method was based on HPLC separation on a reversed-phase C18 column using a mobile phase consisting of phosphate buffer: acetonitrile (40:60, v/v) at a flow rate of 0.8 ml/min. UV determination was achieved at 240 nm. The analytes were subjected to various forced degradation following ICH guidelines. Degradation of metformin was observed under acidic, basic, and peroxide stress. Separation of all the degraded products was achieved. Method was linear in a concentration range of 100-700 µg/ml for metformin and 20-140 µg/ml for gliclazide. The limit of detection and quantitation for metformin was 43.59 µg/ml and 132.09 µg/ml and for gliclazide 8.96 µg/ml and 27.15µg/ml. According to ICH guidelines, the developed method was validated.
