Modern phytochemical analysis in terms of fingerprinting of different bioactive phytoconstituents in the roots of Leptadenia reticulata and whole herb of Cocculus hirsutu belonging to the family Ascelpidiaceae and Menispermaceae respectively. High-performance liquid chromatography (HPLC) analysis of roots of Leptadenia reticulata and whole herb of Cocculus hirsutus were carried out to develop the fingerprint and verify the presence of standards such as Gallic acid, Catechin, Rutin, Quercetin and Kaempferol in the plant extract. RP-HPLC was performed on an Agilent 1260 Infinity equipped with Inertsil ODS 3V (250 mm x 4.6 mm), 5 µm as a stationary phase, by injecting 20 µL of the sample (1µg/mL) at the flow rate 1.5 mL/min and temperature 30⁰C using mixture of Mobile Phase A consist of 1 % Ortho phosphoric acid in water and mobile phase B consist of acetonitrile (20:80) with 60 min run time and the spectra were measured at 364nm using UV detector. According to ICH guidelines, an easy, cost-effective, accurate, and precise RP-HPLC method for simultaneously determining Remogliflzin and Teneligliptin was developed and validated. The drug’s quantity was determined using a photodiode array (PDA) detector. The column was run in isocratic mode with mobile phase acetonitrile and ammonium phosphate buffer at a 40:60 ratio. The flow rate was one milliliter per minute and effluent was measured at 245 nm. The retention times were 2.719 and 7.337 min for Remogliflzin and Teneligliptin respectively. The injection volume was 10µl. as per ICH guidelines the method was validated and the method was found to be linear in the range of 25-150µg/ml for Remogliflzin and 2.5-15µg/m for Teneligliptin. Natural products are becoming increasingly important as alternative medicines and a source of pharmacotherapeutics either directly or as raw materials from which more or less complex chemical structures proven biological activity are isolated. Houttuynia cordata Thunb. is one of the important drugs, which is traditionally used as anti-inflammatory, diuretic, anti-pyretic and detoxicant. In Indo-china, the entire herb is considered cooling, resolvent and emmenagogue. In the present study, the pharmacognostic and phytochemical evaluation of aerial parts of Houttuynia cordata Thunb. (F: Saururaceae) was performed as it is helpful for the standardization and authentication of medicinal plants. The microscopical study showed the presence of blunt-ended multicellular trichomes, lignified fibres, spiral xylem vessels, pitted xylem vessels, striated cuticles, fragments of multicellular trichomes with pointed ends, and cluster crystals of calcium oxalate. A novel, simple, accurate, precise, robust High-Performance Liquid Chromatography method was developed to simultaneously estimate reserpine and piperine. This method was applied to standardize polyherbal formulation containing these markers as one of its ingredients. The markers were resolved using Prontosil C-18 column, with mobile phase acetonitrile and water containing 0.05% ortho-phosphoric acid in the ratio 50:50, flow rate of 1 ml/min and detector set at 225 nm. Retention time of reserpine and piperine were found to be 5.4±0.2 min and 13.5 ± 0.2 min, respectively. Linear response was obtained in the tested concentration range of 1 to 20 µg/ml for reserpine and 10 to 100 µg/ml for piperine, with correlation coefficients of 0.999 and 0.998 for reserpine and piperine, respectively.
