Methotrexate (Mtx) is used to treat a number of conditions, including certain types of malignancy. Therefore, the purpose of this study was to investigate the potential protective impact of flavonoid quercetin on toxicity caused by methotrexate using hematological, biochemical, histopathological and oxidative stress approaches. There were four different groups, each group consisting of ten rats. The group 1 was a control, and animals were treated with 0.5 ml of carboxymethyl cellulose. Group 2 and 3 were treated with methotrexate (0.250 mg/kg); quercetin (50 mg/kg) and group 4 were treated with methotrexate (0.250 mg/kg) + quercetin (50 mg/kg). All drugs were given orally via oral gavage once daily for 28 days. At end of the trial, blood samples were collected from all of the animals for hematological, biochemical and oxidative stress parameters estimation. The objective of this research was to design and validate an alternative accurate method for quantitative determination of Amphotericin B. Amphotericin B deoxycholate life-saving drug belongs to the polyene class approved for the treatment of progressive and potentially fatal fungal infections. Amphotericin B was estimated at 382 nm and the percentage recovery was found to be 100.2 ± 0.34. The method was tested and validated for various parameters according to the ICH guidelines. The repeatability (inter-day) and intermediate precision (intra-day) precision studies revealed that the method is precise and reliable where all the RSD values were less than 2% for the routine analysis of drug. Background: Misleading advertisements in the healthcare sector, particularly concerning Ayurveda, Siddha, Unani, and Homeopathy (ASU & H) products, pose significant risks to public health. Despite existing regulations, the prevalence of such advertisements remains a concern, necessitating comprehensive strategies for mitigation. Methods: This cross-sectional analysis examines misleading advertisements reported by the Siddha Regional Research Institute (SRRI), Puducherry, functioning as a Peripheral Pharmacovigilance Centre for Siddha. Data from August 2018 to October 2023 were analyzed, focusing on reported instances, categorization by disease system, prevalent advertising channels, and implications. Results: A total of 69 misleading advertisements were reported by SRRI during the study period. Analysis revealed a notable prevalence of Non-Communicable Diseases (NCD) in reported advertisements, with a significant focus on Diabetes Mellitus. Print media, electronic platforms, and other sources were identified as major channels for advertisement dissemination. A new simple, specific, accurate and stability-indicating reversed phase `high performance liquid chromatographic (HPLC) method was developed for the determination of Selexipag using a Grace C18 (250mm x 4.6ID, Particle size: 5 micron) a mobile phase consisting of Buffer (pH-7.4): Methanol 60:40, at a flow rate of 1.0 mL/min and ultraviolet detection at 294 nm. The retention times of Selexipag was found to be 4.296 min. Linearity was established for Selexipag in the range of 20-100 μg/mL with correlation coefficients >0.999. The percentage recovery of Selexipag was found to be in the range of 100.99-101.06%. Stress testing was carried out to demonstrate specificity of the method.
