Aloe vera has been used since ages to manage a number of diseases causing pain, inflammation and fever. However, its effectiveness has not been scientifically certified. NSAIDs & Opioid are used continuously for the pain management. Alternative analgesics are needed to escape from the serious adverse effects of NSAIDs. Our study demonstrated significant analgesic activity of Aloe vera at doses of 200 mg/kg and 500 mg/kg. Materials and Methods: The study was carried out using Wistar Albino rats (150-200gm). Soxhlets apparatus was used to prepare the aqueous extract of Aloe vera [AEAV]. The animals were divided into 4 groups (n=6) receiving different treatments. Hot plate method, acetic acid induced writhing method and tail flick tests were used. The results were subjected to statistical analysis by using one way ANOVA followed by Dunnett’s test. The aim of present study is to improve the aqueous solubility of valsartan by utilizing hot melt extrusion technique (HME). Hot melt extrudates were prepared by novel polymeric matrices such as HPMCAS and Plasdone S630 copovidone at different ratios. The solubility studies of extrudates, showed a significant improvement in aqueous solubility. Gastroretentive floating tablets of valsartan using its extrusion complex with plasdone S630 copovidone were formulated by, direct compression method. The tablets comprised of HPMC K4M, HPMC K15M, as release retarding polymers to control the drug release. Preliminary trials applied to optimize the drug release profile. A 10.5 fold increase in the solubility of valsartan was observed with valsartan: plasdone S630 copovidone complex in the ratio of 1:2. Trial batches of tablets showed best results for formulation F3 (20% HPMC K15M and pregelatinized starch 12%) released 99.41% of valsartan in 20 h, with desired floating lag time (18 sec) and constantly floated on dissolution medium for more than 24 h. Antianaphylactic activity of Withania somnifera was investigated by using rat peritoneal mast cells. The investigation was carried out by presensitization of the rat by using sheep serum and tripple antigen. Sensitization induces mast cell degranulation. The rats which were pretreated with standard drug, prednisolone (10 mg) and different extracts (petroleum ether, methanol and aqueous extract) of Withania somnifera at a dose of 250 mg, 500 mg and 750 mg were observed for the degranulation of mast cells, during the anaphylactic reactions. Treatment with methanolic extract of Withania somnifera at a dose of 500 mg shows beneficial effect on mast cell degranulation in sensitized rats. A simple, accurate, precise and easy spectrophotometric method for the determination of levetiracetam in pharmaceutical formulations using molybdenum blue method was developed by the authors. Aqueous solution of the drug sample on reaction with ammonium molybdate solution and hydrazine sulphate solution on heating for 5 minutes at a temperature of 60 0 C gave a clear, stable and intense blue colored molybdenum blue complex. This molybdenum blue showed a λmax at 823nm. Beer’s law was found to be obeyed in the range 0.02-0.08mg/ml. correlation coefficient was found to be 0.998.
