NON-TARGETED ANALYSIS AND CYTOTOXIC ACTIVITY OF HAMELIA PATENS JACQ.
AbstractIn the present study we have done non-targeted analysis of extract from Hamelia patens Jacq. by GC/MS and estimated the inhibitory concentration (IC50) of the extract in three human tumor cell lines MCF-7, H-460 and SF-268. Hamelia patens is an herbaceous perennial plant of family Rubiaceae. Parts of the plant are used in Peruvian and Mexican folk-medicine. Dried leaves of Hamelia patens were extracted for alkaloid. The extract was named HPAE and the yield was 1.76% (w/w). Non-targeted GC/MS analysis of HPAE was performed using Shimadzu QP 2000 GC, equipped with a ULBON-HR-5 capillary column and mass spectrometer as detector. The cancer cell lines were obtained from National Centre for Cell Science, Pune. In-vitro cytotoxic activity of HPAE on cancer cell lines were conducted by MTT assay. The GC/MS analysis of the HPAE enabled the identification of 44 compounds and IC50 of HPAE in MCF-7, H-460 and SF-268 were found to be 8.42 ± 1.16, 90.40 ± 18.48 and 91.47 ± 19.74 µg/ml, respectively. The HPAE was found to be a potential cytotoxic agent against the above cell lines.
