In this study, the in-vivo toxicity of Centella asiatica & Evolvulus alsinoides were evaluated by acute and sub-acute toxicity assays according to the guidelines of OECD 423 & 407 respectively. For LD50 evaluation, a single dose of hydro-alcoholic extracts of both plants was orally administered to Swiss albino mice at doses of 200, 400, 800, 1600 and 2000 mg/kg. Then the animals were observed for 72 h. For acute toxicity evaluation, a single dose of hydro-alcoholic extracts of both plants was orally administered to mice at doses of 300, 600, 1200 and 2000 mg/kg. Then, the animals were observed for 14 days. In the sub-acute study, the extracts were orally administered to mice for 28 days at doses of 300, 600, 1200, and 2000 mg/kg. Centella asiatica L. Urban (family: Umbelliferae) contains several active triterpenoids which have been reported to induce type I collagen which decreases substantially in photoaged skin. Centella asiatica extract is highly soluble in water but have low absorption into the skin. To overcome this, water in oil nanoemulsion loaded with Centella asiatica extract have been formulated, optimized and evaluated as the potential delivery system for treating photoaging. Nanoemulsions were formulated by the oil phase titration method and optimized by constructing pseudo-ternary phase diagrams. The prepared nanoemulsions were subjected to thermodynamic stability testing. Those that passed these tests were characterized for droplet size, polydispersity index, zeta potential and droplet morphology by Cryo-SEM. The prepared nanoemulsions were in nanometric range (optimized batch: 140.5 nm) with uniform size distribution (PDI: 0.086) and good physical stability (zeta potential: -33.9 mV). Centella asiatica (CA) is a reputed medicinal plant that has been shown to have pharmacological effect on skin wound healing. The aim of the study was done to assess the effects of aqueous extract of CA on human dermal fibroblasts in an in vitro model of wound healing. Dermal fibroblasts were cultured with or without supplementation of aqueous extract of CA. Viability and proliferation of the fibroblasts cells were determined by MTT assay and cell cycle were analyzed by flowcytometry. In-vitro re-epithelization was assessed by scratch assay and migration rate was evaluated quantitatively by image analyzer. Gene expression of type I collagen, type III collagen, fibronectin and SMAA2 were studied via real-time RT-PCR. Supplementation with the aqueous extract of CA inhibited the fibroblasts’ proliferation significantly (p<0.05) at high concentration. This study evaluated the anticonvulsant effect of different extracts of Centella asiatica (CA) with particular reference to lipid metabolism in different regions of rat brain. The rats were randomly divided into 4 groups having 6 in each group: i.e. Control group received saline, PTZ-induced epileptic group (60mg/kg b.w op/ 1 day), epileptic group pretreated with chloroform extract (CE), epileptic group pretreated with aqueous (AE) extract and epileptic group pretreated with diazepam (DP; Reference control) (2 mg/kg b.w/ip/ day). The CA extract is administered at the dose of 200 mg/kg body weight orally for one week. The experimental results were observed that the decreased content of phospholipids in the entire brain regions i.e. Cerebral cortex (CC), Cerebellum (CB), Hippocampus (HC) and Pons medulla (PM); total cholesterol, triglycerides and increased content of Lipid peroxidation in epileptic rats.
