Ophthalmic delivery for eye drops which currently cover to 90% of available ocular dosage forms are ideal for the treatment of eye diseases but having limitations of poor therapeutic response and low bioavailability. Objectives: The objectives of present research was to develop of formulation and characterize sustained release in-situ ocular gels containing ciprofloxacin hydrochloride using pH induced gelling polymers for increase drug retention time on eye surface which improved therapeutic response and patient compliance. Methods: In-situ gel formulations prepared by dispersion method using Carbopol-940 alone or in combination with hydroxypropyl methylcellulose (HPMC K4M). Formulations were evaluated for appearance, pH, viscosity, gelling capacity, and drug content and in vitro drug release. The optimized formulation was assessed for sterility and antimicrobial efficacy using disk diffusion technique. Results: The appearance of in situ gels were clear and free flowing in nature however, a viscous clear solution with flow was normal as solution state after gel conversion flow behavior was change. India is the home to the familiar plant Cleistanthus collinus (Roxb.) Benth. ex Hook. F (Euphorbiaceae) has great ethnomedicinal and therapeutic applications. The present study was designed to assess the phytochemical profile and investigate the antifungal effectiveness of leaf, bark and fruit extracts by using the agar well diffusion method. All crude extracts from leaf, bark, fruit and fractions derived also demonstrated significant antifungal activity (20-80%), but the methanol extracts showed a remarkable antifungal spectrum; as a result, GC-MS analysis was performed on it. From this GC-MS analysis, 14 chemicals, including alkanes, glycosides, phenols, flavonoids and steroids were identified, while leaf methanol extract has high phytochemical diversity with 9 compounds than bark with 6 compounds and fruit with 5 compounds. 1, 2, 3-Benzenetriol, 3-O-Methyl-d-glucose and Undecane are a few significant molecules with larger area percentages that demonstrated biological value. Phyllanthus reticulatus Poir., of family Euphorbiaceae was an important therapeutic plant in the traditional medicine, and gaining favour in modern medicine as a plant with varied pharmacological properties. This sericeous shrub is also known as Kirganelia reticulata is widely distributed in India, Thailand, China, Africa and Malaysia. Mouthwash, gargles, mouth spray were formulations were modified using leaves, fruit and stem extract of the plant. The extract of Phyllanthus reticulatus Poir., were studied to have antibacterial, antimicrobial as well as anti-inflammatory activity. Various qualitative tests were performed to authenticate the plant and its property. Presences of alkaloids, phenolic compounds, tannins, coumarins in extract were indicated by the phytoconstituents and UV spectral tests. The extract was found to be stable at acidic state.
