Pectinases are the pectinolytic enzymes that constitute an important component of the plant cell wall and of wide use with their target as pectins. Pectinases cover three different enzymes: Pectin-esterases (PE) act upon ester bonds in pectin breaking them into simpler forms, Polygalacturonases (PG) target the bonds between the galacturonosyl residues, and Pectate lyases (PL) target the alpha 1,4 glycosidic bond. These enzymes can be biosynthesized with the help of various microbes, including bacteria and fungi, using biological waste as a source of nutrients, carbon, and energy. These are commercially important because of their extensive use in the food and textile industry, clarification of beverages, maceration of plant tissue, treatment of wastewater, pulp liquefaction, textile industry and plant fiber processing, coffee, tea and oil extraction, de-gumming plant fibers, and improvement of chromaticity and stability of red wines. This review mainly aims at the pectinase’s, their types, mechanism of action, and the substrates on which they act. It also gives a broad view of the applications of these enzymes in...

The focus of the current article is to prepare the chrono-modulated dual release formulation using various optimization techniques. The conventional formulations as the Controlled Release Drug Delivery System (CRDDS) are not effective in the chronomodulated formulation. The advancement in research and development in CRDDS leads to the formulation of chronomodulated formulations. Dual release tablets are those in which two drugs are given in a single tablet in different layer as to avoid the incompatibility problem or single drug is given in two different layers as maintenance dose and loading dose. The formulation is fruitful in various diseases such as diabetes, arthritis, asthma, etc., and provides compliance to the patient as a patient has no need to wake up during night hours to take the mediation. The Quality by Design (QbD) techniques is used to make the formulation as much as safe and efficacious. By applying the QbD, product variability is reduced and leads to quality improvement, risk reduction, and production enhancement. In conclusion, chronomodulated drug delivery system using QbD...

An amyotrophic lateral sclerosis is a group of progressive neurodegenerative disorders of motor neurons that leads to weakness, muscle atrophy, progressive paralysis, and respiratory insufficiency with a life anticipation of only three years after the onset of disease symptoms. It impairs the functions of both upper and lower motor neurons. The specific mechanism for the disease remains largely unidentified. At present, there is no specific treatment to cure or reverse the progression of the disease. ALS is generally diagnosed based upon the medical history and signs and symptoms; however, it is quite difficult to diagnose the disease in its initial stages. Several studies indicate that various predominant disease mechanisms such as oxidative stress, excitotoxicity, mitochondria dysfunction, aberrant protein homeostasis, defective RNA processing, formation of cytoplasmic inclusion, endoplasmic reticulum stress, endosomal dynamic dysfunctions, protein aggregation, neuroinflammation, oligodendrocyte degeneration, and microglial activation are involved in the pathophysiology of ALS. The various gene mutations (c9orf72, TARDBP, SOD1, and FUS) are also associated with disease mechanisms, so ALS is a complex genetic disorder...

Cancer is one of the most dreaded diseases across the globe. Drug resistance and adverse effects of various classical chemotherapeutic agents are the major challenges for cancer therapy. Cancer treatments with synthetic drugs are associated with severe side effects. Synthetic drugs address symptoms caused by diseases; on the other hand a plant-based medicine usually directs towards aiding body’s own healing process as it is in sync with nature. Natural products and their derivatives can be a promising alternative to existing cancer therapy. Several phytoconstituents have been identified as a valuable source of clinically important anticancer agents. Novel chemical entities and compounds of natural origin have been explored for efficient anticancer activity. Altholactone, a styryl lactone primarily found in Goniothalamus sp., exhibits cytotoxic and antitumor activities. Therefore, an attempt has been made to summarize altholactone analogues. Its structure-activity relationship (SAR) is suggested as a potent anticancer drug candidate, which may provide a structural framework to design and develop a novel anticancer drug in the...

The COVID-19 pandemic has created a global health crisis posing an unprecedented public health emergency. The number of deaths and people being infected is increasing daily throughout the globe. This situation is much more severe due to possible devastating situations because of several social and economic factors. Effective management to address this infection is still evolving, and attempts are being made to integrate traditional interventions along with standard of care. Ayurveda and yoga can be proved excellent results in cure of Covid-19. It has also been accepted by ICMR in its latest guidelines; they have showcase importance of Ayurveda and yoga in treatment of covid-19. In view of this, an attempt is made to conglomerate a few important herbs whose chemical constituents must be tested for their anti-viral potential against Covid-19 infection. Many of the ayurvedic herbal preparation proved its worth as an immune booster in treatment of...
