Most of the Bio-medical wastes got dumped in highly eminent aquatic sources. To control this pollution due to biomedical wastes, the current study focused on the development of hydrogel bead nanocomposite incorporated with the adsorbent and nanoparticles along with biopolymer. The formed hydrogel bead nanocomposite acts as an effective adsorbent of infectious microbes, heavy metals and pollution causing materials. The conjugation efficiency of the activated tea dust, zinc oxide nanoparticles, and bioconjugated system was analyzed using UV-Vis spectroscopy which shows the maximum absorption at 330nm, 379nm, and 580nm in the conjugation system. Further, this was analyzed using FTIR shows good functional groups stretching, Dynamic Light Scattering analysis depicts the particle size as 5.203µm and has good stability. The effectiveness of the hydrogel bead nanocomposite on the wastewater sample was characterized through solubility test and heavy metal adsorption ability. Two-dimensional (2D) culture systems are indispensably used as in-vitro tools to improve our perception and understanding of cell biology, tissue engineering, tissue morphology, mechanisms of diseases, and drug action. However, the conventional 2D culture systems suffer from limitations such as alterations in cell morphology and lack of interactions between extracellular and cellular environments. As a result, three-dimensional (3D) cell cultures have received remarkable attention in studies such as drug discovery and development; the sandwich model being one of the unique cellular models widely used for analyzing tissue metabolism and functions. In this study, we devised a novel Gel Casting Tool (GCT) for in-vitro toxicity analysis. Liver cells need complex 3D micro-architecture to express tissue-specific functions and were suitable for this study. Reverse Phase High-Performance Liquid chromatography is a widely and commonly used analytical method for qualitative and quantitative analysis. A simple, accurate, economical, rapid, and precise RP-HPLC process has been proposed and validated for the determination of esomeprazole in immense bulk and dispensed pharmaceutical drug form. The RP-HPLC separation was achieved on Symmetry ODS RP C18, 5 µm, 15 mm × 4.6 mm i.d. column using mobile phase Phosphate buffer (pH=3.6): ACN = 50:50 pH Adjudged with Orthophosphoric acid at continuance effusion of 1.0 ml/min at ambient temperature. The retention times were observed at 3.797 min for esomeprazole. The calibration plot is linear beyond the concentration range 0-50 μg/ml for esomeprazole. Quantification was achieved with UV detection at 246 nm over the Beer-Lambert’s range. The aim of the present study was to characterize the petroleum ether, chloroform, and ethanol extracts of rhizomes of Curcuma leucorrhiza Roxb and to evaluate their biological activities such as anti-oxidant, anti-bacterial, and anti-fungal activities. Sequential solvent extraction was performed by Soxhlet apparatus using petroleum ether, chloroform, and ethanol, and respective extracts were named as CL-P, CL-C and CL-E. Antioxidant activities of the extracts were evaluated by DPPH, ABTS, and lipid per oxidation assays. Antibacterial and antifungal activities were evaluated against Gram-negative bacteria Escherichia coli (MTCC 739), Gram-positive bacteria Bacillus subtilis (MTCC 121), Micrococcus luteus (MTCC 106), Mycobacterium smegmatis (MTCC 6), fungal plant pathogen Rhizoctonia solani (MTCC 4633) and fungal animal pathogen Aspergillus niger (MTCC 1344) using agar well diffusion method with some modifications. Phytochemical analysis showed that solvent extracts of Curcuma leucorrhiza rhizome contained polyphenol, phenols, quinones, flavonoids, terpenoids, carbohydrates, glycosides, cardiac glycosides, coumarins and alkaloids. Background: Psoriasis is an immune-mediated inflammatory disease characterized by excessive growth and abnormal differentiation of keratinocytes. Objective: In the present research work, an attempt has been made to formulate and evaluate gel containing cyclosporine, aloe vera and tree tea oil. Methods: The gel was prepared by a conventional method and evaluated for various parameters viz., physical appearance, pH, drug content, viscosity, spreadability and in-vitro diffusion studies, In-vitro skin retention studies and stability studies. The prepared optimized gel was further compared with marketed formulation; all the formulations were good in appearance and had good homogeneity. Results: The optimized gel is shown spreadability co-efficient is 5.8 g cm/sec. The gel has shown 95 ± 0.32% releases in 6 h when evaluated for in-vitro diffusion studies. The biological evaluation of the optimized gel has shown 47.98 % drug activity, indicating good efficacy of the formulation against psoriasis.
