ABSTRACT: Fifteen soil samples were collected from different locations near Nagpur i.e. from Nagpur, Bhandara and Chandrapur. Air drying and pre-treatment of soil samples with CaCo3 (1%) were found to be most effective for isolation of actinomycetes. Five different selective media were used for the isolation of actinomycetes and the best growth was observed in yeast malt extract agar medium with 1 mg/ml cycloheximide as antifungal agent and by using double layer agar technique for isolation. Significant differences were found between the counts of actinomycetes on different media. Total 78 actinomycetes species were isolated. More number of actinomycetes were isolated from Nagpur region (32 x 10 cfu/g) as compared to Bhandara (28 x 10 cfu/g) and Chandrapur (18 x 10 cfu/g) regions. Out of 78 isolates 23 isolates showed positive results in primary screening. Maximum isolates showed activity against B. subtilis, B. cereus, S. aureus and E. coli. About 6 extract (methanol, acetone, hexane, ethyl acetate, hexane-ethyl acetate, aqueous extraction) from the leaves of Mangifera indica was extracted. Hexane-ethyl acetate was characterized by Gas Chromatography- Mass Spectroscopy. Ten constituents from 10 peaks were identified. Terpinyl acetate (5.80%) and phytol isomer (5.12%) are as the major constituents and the minor constituents like oxirane (3.57%), sabinene (3.24%), beta-pinen (3.34%), beta-myrcene (3.23%) , cymene (3.68%), alpha-limonene (2.82%), eucalyptol (1,8-cineo (4.71%), 1,3-benzodioxole, 5-(2-, (3.68%) were identified. The antimicrobial activity of different extract was tested against human and plant pathogenic bacteria. Hexane-ethyl acetate extract showed significant role on inhibiting almost all tested pathogenic organisms. Objective: Dental caries is a common oral infection prevalent in developing countries. Streptococcus mutans, a commensal bacterium of the oral cavity, plays a major role in the etiology of caries and root canal infection. Sodium hypochlorite and Chlorhexidine are used in the prevention of caries and as root canal medicament though they cannot be used in combination. Study Design: Different concentrations of passion fruit pulp extract were evaluated to measure the most effective way against Streptococcus mutans. For this bacterial inhibition zones were measured for each concentration in millimeter and compared. Results: The most effective concentration of passion fruit pulp extract was 40% to 45% against Streptococcus mutans.
