Parkinson’s disease (PD) is the second most prevalent type of neurodegenerative disorder and is a chronic, progressive condition. In the current investigation, we used a rotenone (ROT)-developing rat model of PD to assess the neuroprotective efficacy of ethanolic Withania somnifera root extract (EWSR) in comparison to the known neuroprotectants i.e., Doxycycline (Doxy), and ellagic acid (EA) individually and in combination (EWSR-Doxy-EA). Male Wistar rats were given ROT at a dose of 2.5 mg/kg body weight daily for 4 weeks to develop a PD in-vivo model. Once the PD model is confirmed, the rats were treated with individual neuroprotectants for next 5 weeks and kept under observation for next 7 days for reversal of PD. ROT administration has altered behavioural parameters (Cognitive, motor dysfunction, sniffing, grooming, raring, crossing, etc.) and neurotransmitter profile (Acetylcholine, epinephrine, nor-epinephrine, dopamine, glutamate and aspartate) in rats. Objective: The study’s key objective is to evaluate the Antidiabetic activity of methanolic root extract of Daucus carota in normal, streptozotocin-induced Diabetic, and alloxan-induced diabetic rats. Materials and Methods: Albino Wistar rats were administered at a dose of streptozotocin 50mg/kg /day and alloxan monohydrate 150mg/kg/day i.p. for 30 days. Experimental rats were given Daucus carota root extract in two different doses of 200mg and 400mg/kg/day orally. Results: The Preliminary phytochemical investigation of Daucus carota shown that the presence of sterols, saponins, flavonoids, and triterpenoids. In this study, Daucus carota root extract was administered for 30 days and showed antidiabetic activity in STZ diabetic rats and alloxan-induced rats. Conclusion: The methanolic extract of Daucus carota roots also has potential antidiabetic activity in STZ-Nicotinamide diabetic rats. Diabetes mellitus is a challenging human disorder for medical researchers, and the patients suffering from it are from all age groups and countries. BSL-150 tablets is a polyherbal formulation used for the treatment of diabetes mellitus. It contains seeds of Syzygium cumini, bark of Tinospora cordifolia, leaves of Gymnema sylvestre and seeds of Phyllanthus emblica. These plants’ dried, powdered parts in the same quantities as marketed tablets were mixed to obtain the in-house formulation. Aripiprazole is a biopharmaceutics classification system (BCS) II drug used to treat schizophrenia and bipolar disorders. A fast-dissolving tablet will be an appropriate drug delivery route in these conditions. Therefore, the objective of the work was to convert aripiprazole into a solid dispersion via a spray drying process and further present it as an oral fast-dissolving tablet for improving patient compliance. The solid dispersion of the drug in mannitol from solvent evaporation via spray-drying forms porous amorphous solid dispersions. The spray drying trials were optimized using a custom design approach via Design Expert software version 13. Oral fast-disintegrating tablets containing optimized solid dispersions were evaluated for tableting properties, in-vitro drug release profile, and pharmacokinetic profile compared to aripiprazole.
