The present study was designed to investigate the antidiarrheal potential of the methanolic extract of Dillenia indica bark (MDIB). The extract studied for antidiarrheal property using castor oil and magnesium sulphate induced diarrheal model and charcoal induced gastrointestinal motility as well as PGE2-induced enterolooping test in mice. In addition, activities against some causative diarrheal pathogenic bacteria were also determined. At the doses of 100 and 200 mg/kg body weight, MDIB extract significantly reduced the frequency and severity of diarrhea in test animals throughout the study period and also showed a significant (p<0.001; p<0.05) reduction in the gastrointestinal motility in charcoal meal test as well as PGE2-induced intrafluid accumulation. MDIB extract also displayed strong antibacterial effect against some diarrhoic pathogenic bacteria and highest activity was found against Escherichia coli. The degradation behavior of Febuxostat was investigated under different stress degradation (hydrolytic, oxidative, photolytic and thermal) conditions recommended by International Conference on Harmonization (ICH) using HPLC and LCMS. Febuxostat was found to degrade significantly in acidic and alkaline conditions as well as in neutral hydrolysis. The drug was stable to dry heat, photolytic degradation and under oxidative condition. Resolution of drug and the degradation products formed under different stress studies were successfully achieved on a C-18 column utilizing Methanol- water (with 0.02%v/v TFA) in the ratio of 95:5 and at the detection wavelength of 315 nm. The method was validated with respect to linearity, precision, accuracy, selectivity and specificity. The degradation kinetic of Febuxostat in acidic condition at different temperature was studied. Capsaicin, the pungent ingredient of red pepper, is used topically to treat different types of neuropathic pain, in rat model of vincristine induced neuropathic pain we tried to investigate the effect of capsaicin on sciatic nerve through electrophysiological and histopathological studies. We found that treatment of animals with vincristine results in significant decrease in sciatic nerve conduction velocity and degeneration of the nerve fibers, where combined treatment of vincristine and capsaicin showed highly significant decrease in sciatic nerve conduction velocity and degeneration of the nerve fibers compared with that treated with vincristine only. In new trial, we tried to investigate the effect of direct capsaicin titration on sciatic nerve fibers that results in nearly abolishment of nerve conduction velocity. Decrease in blood glucose level test with the use of oral glucose tolerance method from Avocado leaves was carried out. Swiss Webster male mice were divided randomly into five groups. They were negative control, positive control (Glipizide 0.013 mg/20 g BW) and three test groups treated with dosage of avocado leaves ethanol extract 0.490 g/kg, 0.980 g/kg and 1.960 g /BW respectively. Experiment was begun with feeding the mice with the test solution followed by feeding glucose solution (1.5 g/kg BW) 30 minutes later. Blood glucose levels were assessed using glucometer kit, from zero to 3 hours, at ½ hourly interval. These results analysed by one way ANOVA showed there were significant difference (p<0.05) between Ethanol extract of avocado leaves treated with mice (0.490, 0.980 g/kg BW) and control groups.Furthermore, when Tukey’s test was performed, avocado treated mice (1.960g/kg BW) reduce glucose level to 64.27%.
