Four simple, rapid, precise, economical and accurate spectrophotometric methods have been developed for simultaneous analysis of Metoprolol succinate and Telmisartan in their combined dosage form. Method 1, First derivative simultaneous equation method (Vierodt’s method). It employs formation and solving of simultaneous equation using two wavelengths 230.2 nm (λmax of Metoprolol succinate) and 237 nm (λmax of Telmisartan) in first derivative spectra. Method 2, First derivative Q-Absorbance equation method. It involves, formation of Q-absorbance equation at 231.8 nm (isoabsorptive point) and 237 nm (λmax of Telmisartan) in first derivative spectra. Method 3, Absorbance correction method, involves measurement of absorbance at 296.6 nm for estimation of TEL and measurement of corrected absorbance at 223 nm for estimation of MET. Background/Aim: Endosulfan is a pesticide of organochlorine group uses by 55 % of farmers in Bihar. Present study aims to illustrate effect of Endosulfan on biochemical, hormonal and sub cellular anomalies of spermatozoa of mice and their restoration through root extract of Withania somnifera. Materials and Methods: Experimental mice were administered with endosulfan for eight weeks followed by eight weeks administration of Withania somnifera. Results: Endosulfan administered group show degenerated mitochondria and plasmamembrane. Degenerated microtubule were also observed with rudimentary central hub and dynein arm. While eight weeks Withania somnifera 1000 mg/kg/b.w/day administered group show greater degree of sub cellular restoration on mitochondria and nuclear membrane. Microtubules were almost normal in structure. Lipid peroxidation level were also restored toward normal after ashwagandha administration. Lupeol, a pentacyclic triterpene, possesses diverse pharmacological and biochemical activities including anticancer and antioxidant effects. Abnormalities in the status of glycoconjugates and lipids in the cell results in malignant transformation. The aim of the present study was to investigate the protective effect of lupeol on cell surface glycoconjugates and lipids abnormalities during 7,12-dimethylbenz[a]anthracene (DMBA)-induced hamster buccal pouch carcinogenesis. Oral squamous cell carcinoma was developed in the buccal pouches of golden Syrian hamsters by treating with 0.5% DMBA in liquid paraffin, three times a week, for 14 weeks. The status of glycoconjugates and lipids were measured using specific colorimetric methods. We observed 100% tumor formation with marked abnormalities in the status of glycoconjugates and lipids in hamsters treated with DMBA alone. Oral administration of lupeol at a dose of 50mg/kg bw, completely prevented the formation of tumors and restored the status of glycoconjugates and lipids in hamsters treated with DMBA. Churna preparations are an important and widely used form of Ayurvedic herbal formulations in India.
