PCOS (Polycystic ovarian syndrome) is a prevalent female endocrine disorder with global occurrence ranging from 6-26%. It accounts for the majority of anovulatory infertility in women of reproductive age due to enlarged ovaries with small cysts formation, which the National Institute of Health has reported as a cause of 30% of infertility cases in females worldwide. Anovulatory infertility is generally treated with ovulation induction therapy using drugs to promote and facilitate ovulation. However, these medications’ varying success rates and numerous adverse effects compel the search for more promising and safer alternatives. In this research study, 10 ovulation induction agents i.e., Tamoxifen, Metformin, Clomiphene, Pioglitazone, Letrozole, Rosiglitazone, Anastrozole, Prednisone, Dexamethasone, and Spironolactone, were selected to analyze their pharmacokinetics, drug-likeness, bioactivity profile, and toxicity profile through the in-silico approach. Objective: This dissertation work aimed to prepare Levocetirizine HCL (LCH) nasal in-situ gel, a second-generation antihistamine, to improve its retention time, sustain release, enhance the bioavailability, and avoid first-pass metabolism. Process: The in-situ nasal gel was prepared to consume Carbopol as a gelling agent, Hypromellose (HPMC) as a thickening or thickness-imparting agent, and buffering agents to maintain pH. An assessment was conducted to determine the solubility, pH, rheological characteristics, consistency of content, adhesive properties to mucous membranes, gel strength, in-vitro release characteristics, the kinetics of release date, isotonicity, and sterility of the sample. Results: All formulations had a clear appearance with good gelling capacity, and drug content was found to be >87%. The pH values of the formulations were found to be acceptable, ranging from 6.2 ± 0.10 to 6.4 ± 0.44, and no irritation of the mucosal membranes was observed. Optimizing the cultural conditions to enhance bioactive metabolite production by Streptomyces rubrus VLK-24 isolated from mangrove sediments as well as characterizing these compounds, has been taken up in this study. For the present study, 10 bacterial strains were isolated from agriculture field soils collected from Guntur, Andhra Pradesh, India, and thereafter screened for amylase activity. Among them, the strain DS5 showed a maximum zone of clearance on the starch hydrolysis test was selected for this study. Identification of bacterial strain DS5 was confirmed by 16S rRNA sequencing analysis. The phylogenetic tree constructed on the basis of 16S rRNA sequences revealed that it clustered with the closest members of Bacillus pumilus DS5 and the sequences were deposited in NCBI Gen bank along with accession number MK430437. Further the effect of incubation period, temperature, pH, carbon and nitrogen sources was optimized. Highest amylase production was found after 48 h of incubation at pH 7.0 and 40 ⁰C temperature. Diabetes mellitus is a complex disorder where oxidative stress plays a crucial role in the development and progression of the disease. In the present work, the rat model evaluated pancreas and liver tissue damage caused by a high-fat diet and diabetic complications. Male Wistar rats were administered with HFD- corn oil orally for 28 days followed by streptozotocin (STZ) injection to induce diabetes on 29th day. The drug treatment groups were given low dose (250mg/kg) and high dose (500mg/kg) of hydroalcoholic extract of Achyranthes aspera (HEAA), and its therapeutic efficacy in HFD + diabetes model was compared with the standard drug metformin. Various antioxidant parameters were measured, including MDA, protein, GSH levels, SOD, and CAT activities.
