Pioglitazone hydrochloride is a novel antidiabetic drug in thiazolidinediones group and it improves insulin sensitivity in insulin resistant patients. One of the major problems with this drug is its low solubility in biological fluids, which results into poor bioavailability after oral administration. The present study is an approach to enhance the dissolution rate of pioglitazone HCl by preparing solid dispersion (solvent evaporation method) using PEG 6000 as carrier in the ratios of 1:1, 1:2, 1:3 and 1:5 respectively. The drug carrier interaction study was carried out by Fourier Transform Infrared Spectroscopy (FTIR). The prepared solid dispersions were characterized for percentage yield, bulk density, tapped density, Carr’s Index, Hausner’s ratio, angle of repose, drug content, in vitro drug dissolution and stability study. The FTIR study suggesting no interaction between physical mixture of drug and carrier. The solvent evaporation was found to be efficient method to obtained good yield solid dispersions with good flow properties. The drug content was found in the ranges of 72.87±0.31 to 85.23±0.22 %. The solid dispersion...

The present study was to evaluate the in vitro cholesterol esterase enzyme inhibitory and in vitro antioxidant activity of the methanol extract of the leaves of Camellia sinensis (L.). Phytochemical screening of the extract shows the presence of flavonoids, phenolics and terpenoids. The extract shows ability to inhibit the enzyme with IC50 (82.46±0.74µg/ml) whereas that of standard, Orlistat (24.15±0.59µg/ml). Antioxidant and free radical scavenging activity were also assessed by using the methods, IC50 values for Nitric oxide radical scavenging activity (396.83±0.83µg/ml), whereas for standard curcumin (260.38±0.66µg/ml), hydroxyl radical scavenging activity (47.04±2.26µg/ml) and for quercetin (70.99±1.31µg/ml). Moreover, the extract was found to scavenge the superoxide with 50% inhibition at 308.17±23.25µg/ml and standard ascorbic acid at 225.08±2.44µg/ml, IC50 for ferrous chelating ability assay (44.12±4.63µg/ml) and of ascorbic acid (47.25±.89µg/ml). Total content of flavonoids present in 1mg of extract was 19.8±0.11 µg quercetin equivalents/mg. Results indicated that the extract shows potential bioactive compounds which might have a beneficial impact on diseases related to cholesterol synthesis and showed potential antioxidant and free radical...

Eclipta prostrata Linn. (Asteraceae) popularly known as “Bhringaraj” is a very useful medicinal plant in India. The aim of the present study is to evaluate the anti-inflammatory activity of ethanolic extract of leaves of Eclipta prostrata. The anti-inflammatory effects were investigated by employing acute inflammatory model; Carrageenan induced paw edema and Sub-acute inflammatory model; formalin induced paw edema in rats. The ethanolic extracts at a dose of 200mg/kg and 400mg/kg body weight showed significant anti inflammatory activity in both the models in a dose dependent...

Liver is a major metabolic organ affected by various toxins and chemicals. So the potential therapeutic agent is needed for the treatment of liver diseases. Natural products are the best source of remedies for the treatment of all diseases. The aim of our study was to investigate the effect of leaf extract of Coccinia indica against Alcohol combines with CCl4 and Paracetamol induced hepatotoxicity. The effects of oral treatment with Coccinia indica (CI) leaf extracts (100mg/kg and 200mg/kg for 7 days) were studied on hepatic damage induced by alcohol (40% alcohol 2.0ml/100g, p.o. for 21 days) and CCl4 (0.1ml/kg, s.c. on 20th day) and also with paracetamol ( 750mg/kg ip.) in rats. Biochemical parameters in serum like glutamate oxaloacetate transaminase (SGOT), total Billirubin (TB), Alkaline phosphatase (ALP) and total proteins (TP) were estimated to assess the liver function. Alcohol – CCl4 and paracetamol treatment produced an increase in SGOT, ALP, Total billirubin and decrease in total proteins indicating the liver damage. These effects were progressively reduced (SGOT, ALP and Total billirubin)...

A simple and cost effective spectrophotometric method is described for the determination of febuxostat in pure form and in pharmaceutical formulations. The drug was highly soluble in methanol so it was selected as the solvent system for the drug. This ensured adequate drug solubility and maximum assay sensitivity. The linearity range for febuxostat at its wavelength of detection of 315 nm was obtained as 0.2–15 µg/ml.The linear regression equation obtained by least square regression method, were Y =0.0802.X + 0.0036, where Y is the absorbance and X is the concentration (in µg/ml) of pure drug solution. The absorbance was found to increase linearly with increasing concentration of febuxostat, which is corroborated by the calculated correlation coefficient value of 0.9999. The limit of detection and limit of quantification was found to be 0.1743 µg/ ml & 0.5281 µg /ml respectively. The validity of the described procedure was assessed. Statistical analysis of the result shows high accuracy and good precision. The proposed method was successfully applied to the determination of febuxostat...
