Physical interactions between proteins are cardinal to biological processes. To restore their function, proteins must interact with one another. Hub proteins are densely interconnected proteins with a wide range of biological significance and are also to blame for several illnesses, including cancer, autoimmune disorders, and neurodegenerative diseases. The most prevalent of these is Parkinson’s disease. SNCA, alpha synucelin is a hub protein that is the main cause of this disease; along with this DJ 1 and Parkin also responsible for Parkinson’s disease. Through this study, we performed a molecular-level comparative analysis on hub-proteins of Parkinson’s disease with selected synthetic drugs using the in-silico approach. The binding energy and complex energy identified the better efficacy of the drug. The Lipinski rule of filtering and the ADMET pharmacological parameters were satisfied by seven out of the twelve medicines. Levodopa, among these medications, had superior effectiveness with 4RKW and 5C32 in both approaches. Better Lib Dock scores, binding energies, and complex energies with 4RKW were displayed by safinamide. The molecular dynamic modelling...

With the popularity of biologics, treatment for several inflammatory diseases has improved significantly. Biologics are a group of drugs obtained from biological sources that primarily function by suppressing part of the host immune system. However, this process comes at a cost, as it has been reported that the use of biologics carry a major risk in the form of tuberculosis. To verify this claim, a meta-analysis of biologic therapy vs. placebo therapy was designed to test biologics of different inflammatory diseases and check for tuberculosis as their side effect. This test included psoriasis, rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn’s disease, and ulcerative colitis. It was found that biologics used in treating these diseases were mainly responsible for the spread of tuberculosis as a side effect. By mining different databases for literature mentioning biologics that qualify clinical trials and checking for heterogeneity for each study, primary measures were tested for each disease to determine the spread of tuberculosis when treated with these compounds. Meta-analyses were conducted for the study...

The current study’s purpose was to develop and test a multi-herbal anti-diabetic quick-dissolving tablet compared to a commercially available metformin tablet. The multi-herbal anti-diabetic tablet contains natural medicines with anti-diabetic properties, such as Curcuma longa, Cinnamomum zeylanicum, Zingiber officinale, and Trigonella foenumgraecum. This mixture contains the botanical extracts Curcuma longa, Cinnamomum zeylanicum, Zingiber officinale, and Trigonella foenumgraecum. MCC is employed as a diluent and sodium saccharin as a sweetening agent in the tablet formulation to attain this purpose. In the formulation of tablets, superdisintegrants such as crospovidone, sodium starch glycolate (SSG), and a blend of crospovidone and sodium starch glycolate were used to improve dissolve rate and disintegration. Weight variation, drug content uniformity, hardness, friability, wetting time, in-vitro dispersion time and in-vitro drug release investigations were performed on the tablets. Every parameter’s evaluation results for the poly-herbal anti-diabetic pill were within acceptable boundaries. The thickness, hardness, weight fluctuation, friability and in-vitro drug release investigations of the poly-herbal anti-diabetic tablet were all found to be within acceptable...

Drotaverine (DRO) is an anti-spasmodic medication inhibiting phosphodiesterase-4, an enzyme that breaks down cyclic adenosine monophosphate (cAMP). PDE4 inhibition causes an increase in cAMP, which causes smooth muscle relaxation. For DRO, especially the HPTLC technique and stability indicating studies through HPTLC method have not been reported properly in any database. The current research focuses only on a significantly improved HPTLC-based exact, accurate, and specific approach for DRO identification in the presence of degradation constituents employing densitometric detection.DRO was spotted in 6 mm bandwidths with a Camag 100L sample syringe (Hamilton, Switzerland) on a silica gel G60 F254 pre-coated HPTLC aluminium plate with a thickness of 250 µm using a Camag® Linomat-IV applicator. The application rate was 0.4 L/s, with a 6 mm separation between the two bands. Linear ascending development was carried out in a 20 cm × 10 cm twin trough glass chamber, which was then saturated with the mobile phase (Toluene: Methanol; 7:3 v/v).The method was found to be linear over the 200-1200 ng/band range. The stability-indicating...

PCOS (Polycystic ovarian syndrome) is a prevalent female endocrine disorder with global occurrence ranging from 6-26%. It accounts for the majority of anovulatory infertility in women of reproductive age due to enlarged ovaries with small cysts formation, which the National Institute of Health has reported as a cause of 30% of infertility cases in females worldwide. Anovulatory infertility is generally treated with ovulation induction therapy using drugs to promote and facilitate ovulation. However, these medications’ varying success rates and numerous adverse effects compel the search for more promising and safer alternatives. In this research study, 10 ovulation induction agents i.e., Tamoxifen, Metformin, Clomiphene, Pioglitazone, Letrozole, Rosiglitazone, Anastrozole, Prednisone, Dexamethasone, and Spironolactone, were selected to analyze their pharmacokinetics, drug-likeness, bioactivity profile, and toxicity profile through the in-silico approach. The results present information about each drug, including its pharmacological competency and level of toxicity. This study may aid in the direction of future research and the development of more effective ovulation-inducing medications....
