Polypharmacy is defined as the “prescription, administration, or use of more medications than are clinically indicated. It can lead to drug-drug interactions and result in adverse drug events. Therefore, this study aimed to determine the prevalence of polypharmacy and factors that independently predict polypharmacy prescription. 200 Type 2 Diabetes Mellitus (T2DM) patients were enrolled and analyzed. The overall prevalence of polypharmacy was 51%, with the average number of drugs prescribed being 4.68 ± 3.18. The odds of polypharmacy prescription increased with the duration of T2DM, with comorbid conditions, and were higher in patients with uncontrolled glycaemic status. At the same time, the other factors, gender, smoking, and alcohol habits, did not reveal any significant association with polypharmacy prescription. Thus the most common predictors of polypharmacy prescription found were duration of T2DM, comorbid conditions, and glycaemic status of the patient. The findings of this study would be helpful in Government programs in developing, evaluating, and implementing interventions for supporting medicines optimization and deprescribing in...

The purpose of the present research was to develop a suitable, simple, precise, accurate, robust and reproducible RP-HPLC method for reliable simultaneous quantification of Hydroxychloroquine sulphate (HCQ) and Nitazoxanide (NTZ) in a synthetic mixture. The samples were assayed by the cyber lab HPLC instrument using a C18 (250mm, × 4.6mm, D., 5µm) –cyber-sil under isocratic conditions. The mobile phase consisted of Phosphate buffer (pH-3 adjusted with 1% OPA): Methanol: Acetonitrile (30:35:35 v/v/v) with a flow rate of 1.5 ml/min. The wavelength for detection was 266nm (Isoabsorptive point). The Retention time of HCQ and NTZ was 2.8 min and 9. 3min, respectively. The method was validated by determining system suitability, selectivity, sensitivity, linearity, inter-day and intra-day precision, accuracy, robustness and stability in accordance with the ICH guidelines. The calibration curve was linear over the concentration range, 2-10μg/ml for HCQ and 5-25μg/ml for NTZ respectively (n=6). The developed chromatographic method proved to be simple, precise, accurate, robust, and reproducible. Moreover, the samples showed stability at room temperature over a period of...

Biopolymers are gaining much interest and are considered appropriate substitutes for petroleum-based plastics. It is used for various applications in the medical, agriculture, and packaging fields. The bacterial strains from river soil samples were isolated and screened for biopolymer production. The objective of the present study was to produce polyhydroxybutyrate (PHB) from the isolated strains under optimized conditions in a shake flask. The optimization was done with the software tool Design expert – Response surface methodology (RSM). The optimum environmental conditions were: initial pH 7.0, Carbon Source 35g/l, and Nitrogen source 1g/l. Under these controlled conditions, biopolymer yield was obtained 2.25g/L. The FT-IR spectra of extracted biopolymer indicate symmetric (O-C-O) presence at 1600cm-1 and asymmetric absorption peak at 1400 cm-1. The TGA result indicates the thermal degradation of the obtained biopolymer was at 106 ℃. The obtained results suggest that these biopolymers may be similar to...

Docking has become an indispensable method for predicting a molecule’s efficacy and toxicity. This technique is of great value in reducing the pain and suffering caused due to toxic drugs when tested in animals. Rheumatoid arthritis and the pain associated with the condition is a curse to mankind. In this research, the molecular property and toxicity predictions and evaluation of anti-arthritic and analgesic activities of L-Ascorbic acid derivatives were carried out using in-silico models. The molecular property and toxicity were predicted using the software called as OSIRIS. Molecular properties of compounds like ClogP, solubility, molecular weight and drug-likeness were evaluated. Toxicity parameters like mutagenicity, tumorigenicity, irritancy and reproductive effect were also examined. The anti-arthritic and analgesic activities were analyzed using CDOCKER. All the designed 10 molecules were docked with Hase enzyme for evaluation of anti-arthritic activity and opioid receptor for evaluation of analgesic activity. The results showed favourable ClogP, solubility, molecular weight, and drug-likeness values and excellent dock scores with toxicity profiles except for C4 and C6. Compound C10...

A reverse phase high-performance liquid chromatographic bio analytical Technique (RP-HPLC) was developed and validated as per ICH 2019, US FDA 2018 guidelines for the quantification of amlodipine besylate, Rosuvastatin and Valsartan in human plasma using RP – C18 column. The mobile phase in composition (acetonitrile: water in the ratio 75:25% v/v, pH adjusted to 4.0 with acetic acid) was pumped and run at a flow rate of 0.8 ml/min, and for monitoring of eluents, a detector was set at wavelength 245 nm. The precipitation method was employed for the separation of the analyte from the plasma. A calibration curve was generated from the response and respective conc of standard solutions in the Concrange of LLOQ to ULOQ. The method was studied and validated per regulatory guidelines for selectivity, specificity, accuracy, precision, and stability study. Statistical data of the calibration curve of these drugs in the biological matrix was found within the prescribed limit. Results of accuracy and precision of quality control samples of these combined drugs were found (104.25...
