The aim of the present study was to investigate the possible role of Nigella sativa L. seeds extract on fluoride, aluminum and their combination induced reproductive toxicity in Swiss albino male mice. Nigella sativa L. known as black seeds is used as a form of traditional medicinal plant. Fluoride and aluminum are omnipresent metals and are tremendously used in industries, pharmaceuticals, and food additives. The combination study was planned as the two metals co-exist in the environment. The exposure of the combination of metals may cause a hazardous effect on different systems of the body, especially the male reproductive system. The experimental design of the study consisted of different groups. Group 1: control, Group 2: NaF treated (10 mg/kg), Group 3: NS with NaF treated (300 mg/kg + 10 mg/kg), Group 4: AlCl3 treated (100 mg/kg), Group 5: Nigella sativa with AlCl3 treated (300 mg/kg + 100 mg/kg), Group 6: NaF + AlCl3 treated (5 mg/kg + 50 mg/kg), Group 7: Nigella sativa with NaF + AlCl3 treated (300...

Purpose: To formulate and evaluate Olanzapine immediate-release tablets using direct compression. Immediate release of drug from the dosage form is required for rapid onset of action and produce an immediate therapeutic effect. Method: Tablets were formulated using two different natural super disintegrants like Fenugreek and Isabghul and in combination by using Response Surface Methodology. The selected independent variables (Y1) Fenugreek and (Y2) Isabghul showed a significant effect on dependent variables, i.e., disintegration time (X1) and percentage drug release (X2). Drug and excipients compatibility studies were done by FTIR studies. Results: No significant drug-polymer interactions were observed in FTIR studies. The prepared tablets were evaluated for weight variation, friability, hardness, disintegration, and dissolution. A formulation containing 2% fenugreek and 5% isabghul was offered the relatively rapid release of Olanzapine when compared with other concentrations employed in this investigation. The formulation with 4.2% Fenugreek and 5% Isabghul was optimized, and the tablets were evaluated. Conclusion: The optimized formulation AOF14 showed the desired level of % CDR 98.43 at the end of...

Background: Depression is affecting around 5% of the population. In the traditional systems of medicine, many plants and formulations have been used to treat depression for thousands of years. Objectives: This study evaluated the synergic effect of Hydro-alcoholic extract of Rhizomes of Nordostachys jatamansi DC Per Se with combination with Imipramine in Wistar, albino rats, and swiss albino mice normal groups. Materials and Method: Animals of either sex were selected and randomly divided into a test group. Jatamansi extract 10:1 and imipramine dissolved in distilled water were used. Animals were tested for forced swimming test, tail suspension test, and locomotor after given test drug. Results: Nardostachys jatamansi DC, when given to rats, showed a dose-dependent increase in the number of rotations during forced swimming tests in rats. Nardostachys jatamansi DC, when given to mice dose-dependent statistically significant decrease in immobility time, swimming time, and climbing observed. When given along with a combination of imipraine it shows a statistically significant difference in the result, confirmed that it could have synergistic...

Introduction: Inhaled Corticosteroids remain the mainstay therapy for asthma. Uncontrolled asthma (UA) dose not respond to the highest inhaled corticosteroids, which remains a challenge for clinicians. Multidrug resistance-associated protein 1 (MRP-1) are molecular “pumps,” which expel toxins and modulate corticosteroids’ pharmacokinetics and may lead to UA. Moreover, their role in UA is still not clear and needs further investigation. Our study evaluated the expression and functionality of MRP-1 in patients with controlled asthma (CA) and UA. Materials and Methods: UA was defined as a lack of control with high dose inhaled corticosteroids along with the use of other controller medications. Whole blood MRP-1 expression and functional activity on peripheral blood mononuclear cells (PBMCs) in UA (n=38), (mean age = 53.53 ± 11.85, male = 18) patients, CA) (n=60) (mean age = 51.65 ± 16.99, male = 31). MRP-1 was analyzed by flow cytometry. Result: Expression of MRP-1 positivity on lymphocyte gated population was significantly higher in UA than CA (p<0.001). Similarly, the functional activity of MRP-1 was significantly increased...

Polyherbal formulation (HF) containing different herbs have been used to treat diabetic patients by ayurvedic practitioners in India. The present study reports the effect of HF alone and with metformin on different rat models of hyperglycemia. HF treatment with HF alone and in combination with metformin elicited a significant hypoglycemic effect on epinephrine hyperglycemia and alloxan diabetic rats. HF treatment also altered the glucose-tolerance curve pattern both in normal and diabetic rats. Further, HF treatment alone and with metformin increased liver glycogen, glucose transfer in the liver, enhanced glucose uptake process in peripheral muscles, and also improved architecture of β-islets of Langerhans of diabetic rats. The combination of HF with metformin elicited a greater degree of hypoglycemic activity, which was reflected both on blood glucose level as well as other biochemical parameters studied. The hypoglycemic effect of HF may be mediated through pancreatic as well as extrapancreatic systems. Further, HF combination with metformin seems to be useful in diabetes mellitus and needs to be investigated on human subjects to...
