The objective of the present study is to develop a colon targeted drug delivery system by using sodium alginate as a carrier for Mesalamine. Matrix tablets containing various excipients and sodium alginate were prepared by wet granulation technique using different binder systems. The prepared tablets were evaluated for hardness, weight variation, drug uniformity, friability, and in-vitro drug release study. The surface of the device of the best formulation was coated with Eudragit S100 to ensure that the device was more pH-dependent and trigger the drug release only at higher pH. The matrix tablet containing sodium alginate as a carrier and hydroxypropyl methylcellulose as a binder was found to be suitable for targeting mesalamine for local action in the colon as compared to other matrix tablets containing different binders. This analytical study aims to find out various phytochemical and components present in the leaf extract by the GC-MS technique. The methanol leaf extract revealed the presence of phytochemicals like saponins, flavonoids, alkaloids, quinines, glycosides, cardiac glycosides, terpenoids, phenols, coumarins, and steroids. Adaptation of marine actinobacteria to extreme climatic conditions such as high salinity, high pressure and high temperature has modified their physiological conditions to survive and elaborate novel bioactive metabolites. Among the different actinobacterial strains isolated from coastal regions of Guntur District, Andhra Pradesh, India, one potent strain with broad-spectrum antagonistic activity was found and identified as Streptomyces rectiverticillatus VJMS-8. Attempts were made to optimize the cultural parameters for an enhanced yield of antimicrobial metabolite production. Production of bioactive metabolites by the strain was high in tryptone yeast extract dextrose broth as compared to the other media tested. The strain utilized galactose and tryptone as good carbon and nitrogen sources for the elaboration of bioactive metabolites. The optimum temperature and pH for bioactive metabolite production by the strain were recorded as 35 °C and 7.0, respectively. NaCl @ 5% supported high pro-duction of bioactive metabolites. The objective of present research work is design and in-vivo evaluation of ibrutinib self-nanoemulsifying drug delivery system (SNEDDS) for the enhancement of oral bioavailability. Box-Behnken design was used for the study and the results analysed using response surface methodology. Ibrutinib SNEDDS were prepared with various oils, surfactants and co-surfactants and tested particle size, PDI, zeta potential, refractive index, drug release and TEM studies. The optimized ibrutinib SNEDDS has the composition of capryol 90, cremophore EL and transcutol HP. Based on the particle size and entrapment efficiency and in-vitro dissolution studies; F2 is identified as optimized formulation with 85% drug released, whereas less than 1% was released from pure drug at end time of 30 min. In-vivo bioavailability data reported in male wistar rats indicate higher concentration of drug in plasma implying enhanced systemic absorption of ibrutinib from SNEDDS formulation.
