Simultaneous estimation of parent and its metabolite(s) in a single method poses a threat when one has to monitor one mass transition while running the other compound on liquid chromatography-mass spectrometer. This would again become critical once interference is observed at the retention time of compound of interest in-spite of its absence in the sample. In this paper, a selective and sensitive liquid chromatography-tandem mass spectrometry method for the simultaneous determination of Raloxifene and its two active metabolites, Raloxifene 4-Glucuronide and Raloxifene 6-Glucuronide in human plasma was developed and validated. The method involves a rapid solid-phase extraction from plasma, followed by reversed-phase chromatography with gradient flow condition and mass spectrometry detection. Validation parameters were selected ranging from 0.016 to 1.187 ng/mL for Raloxifene, 3.073 to 700.558 ng/mL Raloxifene 4-Glucuronide and 0.311 to 124.526 ng/ml for Raloxifene 6-Glucuronide. The mean recovery for Raloxifene, Raloxifene 4-Glucuronide, and Raloxifene 6-Glucuronide found to be 85.10, 86.30 & 87.20%, respectively. The peak concentration of Raloxifene, Raloxifene 4-Glucuronide, and Raloxifene 6-Glucuronide were 0.330 ng/mL, 153.047...

A rapid, fast, accurate, and precise method was developed for the estimation of lenvatinib in human plasma using methotrexate as an internal standard by reverse-phase high-performance liquid chromate-graphy (RP-HPLC). The separation was carried out by using zodiasil C18 (150 × 4.6 mm, 5 m) as a stationary phase and 0.01 N sodium di-hydrogen phosphate (pH: 4.8): acetonitrile in the ratio of 45:55 % v/v. The analysis was performed at a flow rate of 1.0 ml/min using PDA detector at 240 nm. The column temperature was maintained at 30 °C for better separation and resolution. The retention time of lenvatinib was found to be 4.508 min. The % coefficient of variation of lenvatinib was found to be 2.66%. The % recovery was found to be 94.758%. The method was found to be linear between the concentration range of 28-1120 ng/mL (r2 = 0.999). The lower limits of quantification were 28 ng/mL, which reach the level drug possibly found in human plasma. Further, the reported method was validated as per the...

A number of N-(5-p-tolyl-1,3,4-thiadiazole-2-yl) benzamide derivatives were synthesized from aromatic acid. The antibacterial activity of these newly synthesized N-(5-p-tolyl-1,3,4-thiadiazole-2-yl) benzamide derivatives against Gram-positive bacteria and Gram-negative bacteria was also studied using the minimum inhibition concentration method. The synthesized compounds were showed better results for antibacterial evaluation against gram-positive (Staphylococcus aureus, Bacillus subtilis), gram-negative (Escherichia coli, Pseudomonas aeruginosa) and fungal strains (Candida albicans, A. niger) and were found to be good antibacterial and antifungal agents. Most of the derivatives showed significant antibacterial and antifungal activity against standard. Compared to other methods, the advantageous features of this methodology are operational simplicity, including excellent yields and short reaction time. The developed synthetic protocol represents a very simple and easy to handle for the synthesis of substituted N-(5-p-tolyl-1,3,4-thiadiazole-2-yl) benzamide...

This paper investigates the determination of trace metals named Chromium and Lead set down in the soil as well as in the groundwater due to the tanning industries area of Ranipet town, Vellore city. The tanneries discharge untreated tannery effluents, which get mixed with the soil, water of the Palar river, and underground in this area. The determination of metals was performed for the soil that was collected from the agricultural land adjacent to the Palar River, which bears untreated tannery effluents at pre-monsoon and post-monsoon seasons. The soil samples were collected from three different layers of different depths in these agricultural lands. The first layer is the superficial layer (0 – 10 cm); the second layer is 30 – 40 cm, and the other is 60 – 70 cm below from the surface level. The soil is affected by the untreated effluents through the deposition of heavy metals. The metals might be deposited into the plants and vegetables grown on that agricultural soil, which disturb human health. An extreme...

The present study involves the formulation and evaluation of floating in-situ gel containing piroxicam for the treatment of arthritis. In the FTIR and DSC study for compatibility of piroxicam with polymers, there are no significant changes observed so it is confirmed the absence of drug-polymer interactions. The gastric floating in-situ gel was prepared by using Sodium Alginate, HPMC K 200 M & other ingredients. The prepared situ gel of piroxicam was evaluated for pH, gelling capacity, floating lag time, total floating time, drug content, viscosity, in-vitro drug release. An optimization study was carried out by using was 32 factorial designs to find the effect of independent variables, i.e., amount of Sodium Alginate (X1) and HPMC K 200 M (X2) on dependent variables i.e., floating lag time & % CDR. The results showed that all the formulations exhibited a very short floating lag time. Most of the formulations floated within 1 min. All the formulations exhibited a basic pH in the range of 6.7 to 7.9 which is suitable for...
