Silica lipid-hybrid formulations have gained an increased interest in recent times as a means to enhance the solubility as well as dissolution of poorly water-soluble lipophilic drugs. The objective of the present investigation was to design and optimize silica lipid hybrid particles of Canagliflozin to improve its solubility. Canagliflozin is an antidiabetic drug used to improve glycaemic control in people with type 2 diabetes. In this study, we have fabricated a Solid-State Lipid-Based Oral Drug Delivery System as CNZ SLH with liquid lipids like Miglyol 812, Labrasol and Acrysol el 135. Sylloid 244 FP grade highly porous silica was used as adsorbent. The interaction of CNZ-SLH particles was studied using X-ray powder diffraction (PXRD), Differential Scanning Calorimetry (DSC) and FTIR spectroscopy. CNZ-SLH particles prepared with Miglyol and Labrasol enhanced the solubility of CNZ by 19.67 fold. A new analytical method has been developed for the simultaneous estimation of Telmisartan and Hydrochlorothiazide in their pure and pharmaceutical dosage forms by using Fourier Transform Infrared (FTIR) Spectroscopy. This method involves the extraction of Telmisartan and Hydrochlorothiazide from tablets using chloroform and direct measurement in liquid phase mode using reduced path length cell. The spectra were measured in absorbance mode, equipment was configured to collect spectra at 8 cm-1 resolution, and the spectra were collected between 4000 cm-1 and 450 cm-1. The infrared spectra showed different peaks with baseline correction, among which intense, clear and proportionate peaks were selected at 3411 cm-1, and 1309 cm-1 corresponding to the N-H group and asymmetric SO2 group for Telmisartan and Hydrochlorothiazide respectively for quantitative estimation. Beer-Lambert’s law was obeyed over the concentration range of 50-250 μg/ml for Telmisartan and 25-125 μg/ml for Hydrochlorothiazide. All the results were found to be within the limits. The present investigation deals with the development of duloxetine ethosome then loading into the transdermal patch to impart lower drug side effect, enhanced bioavailability, avoiding first-pass metabolism. 32 factorial design was applied to optimize the formulation. Ethanol (X1) and Phospholipid (X2) were taken as independent variable, Responses are vesicle size (Y2), entrapment efficiency (Y1). Optimum desirability was identified and, an optimized formulation was prepared, characterized and loaded into transdermal patch. The transdermal patch was evaluated for drug content, thickness, folding endurance. Ex-vivo permeation study for the prepared patch was conducted and, the permeation parameters and drug permeation mechanism were identified. The percent of alcohol was significantly affecting all the studied responses while the other factors and their interaction effects were varied in their effects on each response.
