The present study was aimed to develop the effective protocol for induction roots and in-vitro regenerated plants from embryo culture of Nothapodytes nimmoniana. Excised embryos were sterilized under the aseptic condition and cultured on L2 medium fortified with different concentration (0.5 – 3 mg/L) and the combination of auxins and cytokinins. Maximum shoot initiation was observed in BAP (2 mg/L) and Kin (1 mg/L) and in a combination of BAP (0.5 mg/L) and NAA (2 mg/L). A higher number of roots were observed when the embryo was inoculated on L2 medium supplemented with NAA (1 mg/L) and 2, 4-D (0.5 mg/L). Whereas, elongated roots were observed on L2 medium fortified with BAP/KIN (0.5 mg/L) and NAA (1 mg/L). The proliferation of multiple shoots from the nodal explants of in-vitro plantlets, when sub-cultured on MS medium supplemented with BAP (2 mg/L), TZD (0.2 mg/L) and L-glutamine (100 mg/L). The obtained plantlets were hardened in soil-rite and then transferred them to a mixture of soil, sand, and manure (1:1:1). In conclusion,...

A series of novel analogues of 2-(5-(1H-indol-3-yl)-1, 3, 4-thiadiazol-2-ylimino)- 5- (substituted benzylidene)thiazolidine-4-one have been synthesized. The structures of newly synthesized compounds were confirmed by FT-IR, 1H-NMR, 13C-NMR and Mass spectroscopy. The synthesized compounds showed significant antibacterial activity against gram-positive bacteria: Staphylococcus aureus (MTCC 3160), Bacillus subtilis (MTCC 2061), gram-negative Escherichia coli (MTCC 1652), Pseudomonas aeruginosa (MTCC 741) and antifungal activity against fungal strains: Candida albicans (MTCC 183) and Aspergillus niger (MTCC 2110). Also, their anti-inflammatory activity was evaluated by using carrageenan-induced rat paw edema method. Compounds 7d and 7h with the methoxy substitution on phenyl ring were found as active derivatives of the series, exhibited 49.86% and 49.88% inhibition respectively as compared with Diclofenac sodium. In-silico molecular docking studies of the synthesized compounds was done on crystal structures of proteins of microbes Aspergillus niger, Bacillus subtilis, Candida albicans, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and cyclooxygenase-2 using GRIP batch docking method of V-life MDS 3.0 software to study their observed activity which revealed a significant correlation between the binding...

Regular intake of carbohydrate diet containing high fructose or sucrose increases adipogenesis resulting in conditions like weight gain, high blood pressure, elevated blood glucose levels, insulin resistance, raised triglycerides, low HDL cholesterol, with metabolic syndrome as a leading consequence. Since intricacies of adipogenesis are of major relevance to human health; present study aimed to evaluate the morphological effects of trigonelline on mesenchymal stem cells, C3H10T1/2 treated with high fructose (equal to that in high fructose corn syrup) in comparison to the adipogenic cocktail. In this regard, cells were given trigonelline alone as well as in combination with an adipogenic cocktail or high fructose to evaluate its potential to affect adipogenesis morphologically. The cells of positive control (containing only adipogenic differentiation media) depicted morphological changes towards spherical structure, a feature definite of adipocytes. Similar observations were seen in cells treated with high fructose (1 mM and 30 mM) but not in cells treated with trigonelline alone (75 μM and 100 μM). However, the cells treated with high fructose along with...

In this study, the synergistic effect of an extract of Azadirachta indica and Curcuma longa were investigated by the agar diffusion method. Different dilutions of the extract were prepared, and antifungal activity of the extracts of Azadirachta indica and Curcuma longa against Candida albicans was measured alone and in combination with fluconazole gel. The zone of inhibition of Azadirachta indica and Curcuma longa was found to be 9 mm to 14 mm and 8 mm to 13 mm respectively in the concentration range of 10-60 µL against Candida albicans. The fluconazole gel was prepared, and various evaluation parameters like; spreadability, pH, viscosity, and drug content were determined. Extracts of Azadirachta indica and Curcuma longa were taken in different strength (10-60 mg/ml) however the concentration of fluconazole gel was kept constant (1%) and zone of inhibition was determined. The maximum zone of inhibition was found with Azadirachta indica in the combination of 60mg/ml and Curcuma longa 20...

The present study was designed to evaluate the muscle relaxant and locomotor activities of petroleum ether (PE), ethyl acetate (EA) and methanolic (MA) extracts of Malpighia emarginata (ME) and Holoptelia integrifolia (HI). The different concentrations were used to evaluate the toxicity in rats and revealed that they are nontoxic up to 2000 mg/kg. From the toxicity study, 200 and 400 mg/kg doses were selected for the pharmacological activities. Muscle relaxant and locomotor activities were done by Rotarod and Actophotometer respectively. The leaves and fruit extracts of PEME, EAME, and MAME with 200 and 400 mg/kg were showed a significant reduction in the time spent by the animals on the revolving rod when compared to the control (P<0.01, P<0.001) at 2 h. The MAHI of leaves and bark extracts (200 and 400 mg/kg) were more significant towards muscle relaxant effect (P<0.01, P<0.001) than PEHI and EAHI (P<0.05, P<0.01) when compared with control at 1 h. Treatment with both doses of leaves and fruit extracts PEME, EAME and MAME significantly (P<0.01,...
