The objective of this work was improvement in the ocular bioavailability of diclofenac sodium by enhancing drug permeation across cornea and promoting drug retention time on ocular surface. Using pseudo ternary phase diagrams transparent regions were identified and nanoemulsions were prepared by self-emulsification method, which were further optimized by emulsification study and drug content. Three best NE formulations were incorporated in 20% poloxamer 407 (thermosensitive polymer solution) to formulate nanoemulsion embedded thermosensitive in-situ ocular gel and further evaluated on the basis of gelation temperature and drug entrapment. Optimized nanoemulsion formulation was evaluated by TEM, particle size analysis and zeta potential. The optimized NE gel formulation changed from sol-gel phase at physiological temperature. Comparison of dissolution profile of developed formulation was carried out with the marketed formulation. The formulated NE in-situ gel showed drug release for a longer duration of time as compared to the marketed eye drops (0.1% w/v Voltaren Ophtha). Anti-diabetic and protective activity of T. dioica aqueous extract in diabetic complications in streptozotocin (STZ)-induced diabetic rats was evaluated. Albino rats (n = 24) were divided into four groups, of six animals each. Group 1 (normal control) and Group 2 (diabetic control) received normal saline (10 ml/kg/day p.o.) whereas Group 3 (Standard, insulin 6 U/kg/day s.c) and Group 4 received 1000 mg/kg/day p.o. of T. dioica extract for 28 days. Extract treated animals showed significant decrease in various biochemical parameters like blood glucose, SGOT, SGPT, creatinine, urea, LDL, Cholesterol and glycated hemoglobin. Bacterial biofilms due to antibacterial resistance are causing serious health issues with respect to infection control, its prognosis and treatment of disease. The current scenario demands need of new and alternative antibacterial compounds for treatment of such infections. The current study highlights the use of polluted river, Waldhuni, as a source of potent microorganisms with antibiofilm properties. 186 microorganisms were isolated from Waldhuni River, and tested for quorum sensing inhibitory activity (QSI) against C. violaceum MTCC 2656. Seven isolates exhibited QSI activity, out of which 1 isolate PB2 was selected for further study based on its high QSI potential. PB2 displayed broad spectrum antibiofilm activity against eight strains of biofilm forming pathogens. The effective minimum biofilm eradication concentration (MBEC) of crude extract of PB2 secondary metabolite (SME) was found to be 500ppm. The extract reduced exo-polysaccharide (EPS) by 94.16%, viability by 90% and biofilm density by 96% in 6h treatment. Rapid, sensitive, specific and validated colorimetric methods have been developed for the quantitative estimation of ambroxol HCl in bulk and dosage form. The current method was developed based on oxidation of ambroxol with Gibb’s reagent (method I) and p-diethyl amino benzaldehyde in toluene (method II). The formed intense colour complex was measured at 537.2 nm and 438.4 nm respectively. Under optimized experimental conditions, Beer’s law is obeyed in concentration range 5 – 30 µg/ml for both the methods with regression co-efficient 0.9994 and 0.9992. Objective: To assess the pattern of adverse drug reactions (ADRs) in a tertiary care hospital. Materials and methods: This study was conducted in VSS Medical College, Burla, Odisha from October 2012 to September 2014. ADRs collected from various clinical departments were compiled and analyzed for distribution of age, gender, department, drug class and organ system. Assessment of causality, severity, and preventability was done using specific scales. Results: The occurrence of ADRs was more in males compared to females in the age group 19-60 years of age. Most of the ADRs were collected from medicine department followed by dermatology and psychiatry. The skin was the most common organ system affected followed by gastrointestinal and central nervous system. Antiretroviral drugs were the most common drugs causing ADRs followed by antimicrobials and antipsychotics.
