The objective of this study was to evaluate and optimize Eudragit concentration and coating level on metronidazole release from granules for colon delivery using 32 full factorial design. Independent variables selected were Eudragit conc. (10, 15 and 20%) and coating level (10, 15 and 20%), the evaluated responses were percent drug release at 5 h, 24 h and time to release 50% of the drug. The dissolution was carried out at different pH media of 1.2, 7.4 and 6.8. The dissolution data revealed that polymer conc. and coating level were having significant role in achieving optimum formulation. Using surface response, contour plot and statistical equations, the optimum formulation consisting of 15% of polymer conc. and 20% coating level was predicted. Experimental data showed that the granular formulations releases no drug at pH 1.2 (simulating stomach) and only 12.85 ± 1.63% at pH 6.8 (simulating small intestine) and controlled at pH 7.4. Endophytic actinomycetes are the potential source for obtaining novel bioactive compounds. Present study evaluates the chemical constituents in Ocimum basillicum and its endophytic actinomycetes. Eleven endophytic actinomycetes were isolated from O. basillicum plant. Among eleven, one isolate was characterized by 16S rRNA and identified as Streptomyces flavoviridis A3WK. A comparative analysis of the FT-IR, GC-MS, antibacterial, antioxidant and anti-inflammatory properties of plant and endophytic actinomycetes extracts was conducted. 12 compounds each of Streptomyces flavoviridis A3WK and O. basillicum were identified by GC-MS. The FT-IR and GC-MS analysis reveals that S. flavoviridis A3WK has more antimicrobial, antioxidant activity and anti-inflammatory properties compared with the results of O. basillicum. Over all the activity of the endophytic actinomycetes was more compared to that of the O. basillicum extracts. Ephedra gerardiana is an important medicinal plant used as Somalata in Indian Ayurvedic system of medicine and as traditional Chinese medicine since several thousand years. Plant is known to contain valuable secondary metabolite such as ephedrine and pseudo ephedrine. These secondary metabolites are utilized for several medicinal purposes. A spectrophotometric and reverse phase high performance liquid chromatography methods has been developed for the estimation of Ranolazine in bulk and pharmaceutical dosage form. For UV spectrophotometric method Ranolazine shows maximum absorbance at 273 nm with the concentration ranges of 20-150 µg/mL. The detection limit and quantification limit were found to be 1.23 and 5.21 µg/mL, respectively. Accuracy was in the range of 99.8-100.2%. Intraday and interday precision was in the range 0.16- 0.38% and 0.50-0.82% respectively. The RP-HPLC separation was achieved by using Phenomenex Gemini C18 (100mm X 4.6mm), 3μm column and buffer: acetonitrile (60:40, v/v), (1.0 mL of triethylamine in 1000 mL milli- Q water, pH adjusted to 6 using dilute o-phosphoric acid solution) as mobile phase. The flow rate was 1.0 mL/min and effluents were monitored at 224 nm. The retention time was 5.08 min. The linearity was in the range of 20-150 µg/mL. The Pharmacological effects of silver nanoparaticle of Stemodia viscosa aqueous plant extract on albino wistar rats was explored. The plant extract 40 ml was added to 10 ml of 10mM silver nitrate aqueous solution and kept at room temperature for synthesizing silver nanoparaticle. Diabetes was induced in overnight fasted mice by single intra-peritoneal injection of 55 mg/kg of Streptozotozin (STZ), freshly dissolved in 0.1M cold citrate buffer, pH 4.5. After five days of STZ administration, blood was collected and plasma glucose levels were determined. For wound healing activity, a control ointment base was formulated without any drug content.
