In the present study, the various extracts viz aqueous, ethanol, chloroform and Petroleum ether of Biophytum sensitivum whole plant were evaluated for the anticancer activity against cancer cell lines. The cervical (Hela) and liver cancer (HepG2) cell lines were used to screen the in vitro anticancer challenging with the B.sensitivum extracts. Concentration ranges from 20 to 100 microgram of B.s crude extracts were screened against Hela and HepG2 cells. The antiproliferative activity of B.sensitivum were performed by 3-[ 4,5–Di methyl thiazol-2-yl]-2, 5-diphenyltetrazolium Bromide (MTT) assay. The cytotoxicity was highly significant in the ethanolic extract and chloroform extract than the aqueous and petroleum ether extract. A new RP-HPLC method was developed for Mephenesin (MEP) and Ibuprofen (IBU) and validated as per ICH guidelines. Good chromatographic separation of Mephenesin and Ibuprofen was achieved by using Agilent C18 column (150mm×4.6mm, 3.2µ p.s). The system was operated at ambient temperature using a mobile phase consisting of Acetonitrile, 0.01M potassium dihydrogen phosphate pH 3.0 (60:40 v/v) isocratically at a flow rate of 0.5 ml/min. Detection was carried out at 212 nm and retention time was 3.4 min and 9.3mins respectively. Linearity was achieved from 10-40 µg/ml for Mephenesin and 2-8µg/ml for Ibuprofen with r2>0.99. The analytical method validation studies were performed as per International Conference on Harmonization-Quality (ICH-Q2 (R1)) guidelines. The antimicrobial effects of I. balsamina seed hexane extract against Staphylococcus aureus, Klebsiella pneumonia, Proteus vulgaris, and Serratia marcescens were determined using the standard technique. As well as the phytochemical properties of the extract on the test isolates were also examined using the standard methods. The phytochemical components of the hexane extract of the I.balsamina seed include falvonoids, alkaloids, phenol, tannin, alkaloids, steroid, saponin, phenol, flavonoids, triterpenes glycosides and carbohydrate. All the test organisms were susceptible to 25mg/ml of the extract. The MIC and MBC of the hexane extract of the I.balsamina against S. aureus and E. coli was 50mg/ml, while that of K. pneumonia and S. marcescens were 75mg/ml and 100mg/ml of the extract respectively.
