Aim: The objective of the present research work was to formulate and compare two different kinds of penetration enhancer in transdermal patch with placebo patch using a drug (Salbutamol sulphate) and compare for in vitro drug release. Study Design: The transdermal patches of Salbutamol sulphate were prepared by solvent evaporation technique using different ratios of Hydroxy propyl methylcellulose (HPMC K15M) and Poly vinyl propylene (PVP K30). The prepared transdermal patches were evaluated on parameters like weight variation, thickness uniformity, moisture content, moisture uptake, folding endurance, tensile strength, drug content, in vitro dissolution studies, in vitro drug release, skin irritation test and stability studies. Maximum drug release was showed by Batch T, and the optimized formulation showed satisfactory...

There is a strong association between elevated heart rate (HR), systemic inflammation and atherosclerosis. We assumed that HR lowering by ivabradine might decrease inflammation in patients with non ST-segment elevation acute coronary syndromes (NSTE-ACS). Objective: Study the effects of ivabradine add on treatment on High sensitivity C-reactive protein (hsCRP) levels in patients with NSTE-ACS. Methods: This prospective, randomized, controlled, study recruited NSTE-ACS patients with HR ≥70beats per minutes. Each patient was randomly assigned to either control or ivabradine groups. The difference between the two groups was the addition of ivabradine (up to 7.5 mg bid) to the standard treatment of NSTE-ACS patients for 30 days in the ivabradine group. Levels of hsCRP were evaluated before and after the study period. The primary outcome was the difference between the two groups inhsCRP reduction. Results: We enrolled 45 patients, twenty three of which received ivabradine. The decrease (%) in HR after treatment was significantly larger inivabradine group than in control group (23.8 (7.3 – 31) vs 4.7 (0 – 22.5) %,...

The inhibitory role of trace and major elements in urinary stone crystallization was investigated in five widely used anti-urolithiatic medicinal plants of North East India. Fifteen elements namely K, Ca, Sc, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se and Br were detected with different concentrations (in ppm level) by Proton Induced X-ray Emission(PIXE) technique while light elements namely F, Na, Mg, Al, P and Cl by Proton Induced Gamma ray Emission(PIGE) technique in the studied plants. Analysis was performed on thick targets (pellets) prepared using the powders of specimens through a series of processing steps. Thick targets of plant based Certified Reference Materials (CRMs) were served as standardization of PIXE and PIGE set...

Quality by Design (QbD) is a philosophy that refines the level of knowledge associated with a product that uses process understanding to deliver a product with the desired critical quality attributes. The objective of this present study was to develop and demonstrate an integrated multivariate approach to develop and quantify the constituent concentrations of saxagliptin drug in its pure and formulated forms. The method was developed using a mobile phase acetonitrile: water (pH3), (20:80 v/v) on an Agilent, TC C18 (250 × 4.6 mm) 5μm column and flow rate 1.0 ml/min which was optimized with help of design expert software and validated according to ICH Q2 guideline and application of this method to different stress condition of saxagliptin. The detector linearity was established in concentrations ranging from 10-90μgmL-1, the regression coefficient was 0.9993. The method fulfilled validation criteria and was shown to be sensitive, with limits of detection (LOD) and quantitation (LOQ) of 0.003 and 0.010?gmL-1, respectively. The percentage relative standard deviations for robustness and ruggedness were observed within...

Migraine is an episodic headache disorder characterized by a combination of neurological, gastrointestinal, and autonomic symptoms. Eletriptan hydrobromide (EHBr) is a second generation triptan drug intended for treatment of migraine headaches. The main aim of the study was to prepare and evaluate elastic niosomes of EHBr (ENEH) and optimise the concentration of non-ionic surfactants and ethanol by ethanol injection method. The ENEH formulations were characterised for vesicular shape, size, zeta potential, entrapment efficiency and percutaneous permeation. The formulations exhibited entrapment efficiencies of 19.21±0.124% to 58.20±0.147%. The optimised formulation was FE2 consisting of Span 80 and Tween 20 prepared by ethanol injection method exhibited vesicle size of 207.2±53.7 nm, zeta potential of 19.5 mV entrapment efficiency of 69.70%, flux of 53.903±0.13 μg/cm2/hr, Q8 of 448.262±0.18 µg/cm2, permeability coefficient of 0.013 cm/hr and lag time of 0.2±0.14 hrs and skin content of 1423±12.4 μg/gm. In vitro and ex vivo drug permeation across rat skin revealed improved drug permeation and higher transdermal flux with ENEH formulations compared to niosomes and drug solution....
