The demand for herbal medicines is growing at a drastic rate due to their improved pharmacological actions, minimal side effects, and cost-effectiveness. The ever-increasing cost of chemotherapy ruined the economic stability of many families. This led to the discovery of herbal medicines for cancer treatment. Tribes of Western Ghats have been using the roots of Apama siliquosa for cytotoxic activity. The methanolic and aqueous root extracts of Apama siliquosa Lamk were used to prepare different concentrations. The main objective of this study was to Assessment of the acute toxicity of ethanolic extract of Polyherbal formulation in Swiss albino mice. In this study, the acute oral toxicity study of ethanolic extract of PHF1 was carried out according to Organization for Economic Co-operation and Development (OECD) guideline 423 and 407 on Swiss albino mice (20–30 g). In the study, the oral dose of 2000 mg/kg of PHF1 extract was administered in the treated group. General behavior, morbidity, and mortality were determined up to 72 h and compared to control group. Further animals were investigated for a period of 3days assess any other toxic effect. Behavioral changes and other parameters such as body weight, hematological parameters, and biochemical parameters were evaluated and compared to the normal group. The focus of the current article is to prepare the chrono-modulated dual release formulation using various optimization techniques. The conventional formulations as the Controlled Release Drug Delivery System (CRDDS) are not effective in the chronomodulated formulation. The advancement in research and development in CRDDS leads to the formulation of chronomodulated formulations. Dual release tablets are those in which two drugs are given in a single tablet in different layer as to avoid the incompatibility problem or single drug is given in two different layers as maintenance dose and loading dose.
