Bombax ceiba is a traditional medicinal plant that is beneficial in the treatment of diuretic, hemostatic, antidysenteric, emetic, antipyretic, astringent, and commonly known silk cotton tree, semal, kapok tree. The aim of this study is to isolate and characterize the biologically active compounds from the ethanolic extract of the leaves. The EtOH extract was subjected to silica gel (60-120 mesh) by glass column chromatography, using gradient petroleum-ether, C6H6, CH3OAc, CH3OH, and purified by preparative TLC (GF254) to yield two pure natural products (taraxerol and β-sitosterol glucoside). The structure was elucidated on the basis of spectroscopic analysis, including 1HNMR, 13CNMR, DEPT-135, COSY, HSQC, and HMBC. The β-sitosterol glucoside shows pharmacological activities such as lowering cholesterol levels, reducing blood pressure, antitumor activity, etc. The glucose moiety is highly effective because it is hydrophilic, which inhibits the entry of cholesterol into the esterification of cholesterol. It is the first time isolated from the leaves of this...

Dieffenbachia amoena is a house plant and known dumb cane. The study was performed based on isolation and structure elucidation of phytosterols from the extract of this plant. The CH3OH crude extract was loaded over the silica gel (60-120 mesh) column, using the stepwise gradient C6H6, CH3OAc, CH3OH. The fractions were further purified by preparative TLC (GF254) to yield compound (1). Compound (1) was characterized by using various standard spectroscopic techniques such as IR, 1HNMR, 13CNMR, DEPT-135, COSY, HSQC and HMBC. Based on the spectral analysis, it was confirmed that the compound (1) was a mixture of β-sitosterol and stigmasterol. Phytosterols have high medicinal importance and play a vital role in reducing blood cholesterol, a high level of blood cholesterol can cause a risk of cardiovascular disease. Cardiovascular disease is the main problem of the whole world and increasing day by day. We have isolated β-sitosterol and stigmasterol first time from the leaves of the Dieffenbachia amoena...

Depression is the most common and spreadable disorder in recent age as people are going through a lot of pressure in their daily work caliber and so many unconditional events. A new approach called derivatization spectroscopy was developed for the anti-depressant agent, marketed formulation of Bupropion hydrochloride, to confirm the interfering substances from the excipients of solid dosage form, in addition to that the stability parameters to assure the analysis time with the minimum usage of the reference standard and sample solution. The present work aims to develop simple, sensitive stability indicating the method to develop a model to estimate BPH in the solid dosage form by using derivative spectroscopy tool to avoid the excipients interferences in the developed assay method. First, made the stock solution of 100 µg/ml from 1000 µg/ml previously prepared solution. The method was validated over the concentration range of 5-50 µg/ml of marketed formulation of Bupropion Hydrochloride. The assay method & derivatization was performed using UV spectroscopy at 252 nm wavelength. The intra-assay &...

In this study we aimed at isolation, identification, and quantification of two phenolic compounds reported for the first time in leaves of Amaranthus hypochondriacus. A non targeted GC-MS metabolite profiling of hexane extracts of amaranth leaves was done which indicated the presence of major phenolic compounds. Two alkylated phenols namely 2,4-di-tert-butyl phenol (A) and 3-(3,5-di-tert-butyl-4-hydroxyphenyl) propanoic acid (B) were isolated for the first time from the hexane extract of A. hypochondriacus leaves. Structure elucidation of both the compounds was done using 1H NMR and purity was checked by GC-MS. Quantitative estimation of purified compounds in leaves extract was carried out using RP-analytical HPLC. The antioxidant potential of the isolated compounds (A), (B) and the hexane extract of leaves were investigated using in-vitro 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radicals scavenging bioassay. Hexane leaf extract (IC50 = 385.34 ± 37.11 µg/mL), 2,4-di-tert-butyl phenol (A) (IC50 = 253.76 ± 24.67 µg ml-1) and 3-(3,5-di-tert-butyl-4-hydroxyphenyl) propanoic acid (B) (IC50 = 174.09 ± 16.89 µg ml-1) exhibited significant free radicals scavenging activity. Thus, our study reports...

Bromelain is a protease present in pineapple fruit and stem. It is reported that commercially available pure bromelain has anticancer, anti-inflammatory and other therapeutic effects. But there are few reports available on the biological activity of bromelain along with peroxidase which is present in pineapple fruit extract. The aim of the present study was to investigate the effect of bromelain with peroxidase from pineapple fruit against lymphoma cell line in in-vitro. Pineapple was purchased from the local market from Tripura. Bromelain isolated along with peroxidase and enzyme activity tested with the substrate. In-vitro cytotoxicity was determined by MTT assay, and apoptosis was determined by fluorescence microscopy, DNA fragmentation assay and by western blot analysis. Fluorescence microscopy analysis and the MTT assay showed significant apoptosis of lymphoma cell after treated with bromelain along with peroxidase in a dose- and time-dependent manner and was more apoptotic than bromelain alone. This was further confirmed by DNA fragmentation assay. Furthermore, western blotting analysis confirmed the mitochondrial pathway is involved in apoptosis of lymphoma...
