Ancient DNA help in better understanding the evolution and migration pattern of humans throughout the world. The present work is a preliminary effort, to extract and amplify the ancient DNA from human bone samples provided by the Anthropological survey of India collected from the Himalayan region in 1952. Many technical difficulties arise during DNA extraction, PCR amplification and sequencing which make it difficult to work and interpret the analyses.Our preliminary result is encouraging, as we have successfully amplified 456 base pair PCR product of mitochondrial DNA hypervariable region I (HVR1) starting from position 15976 to 16431. Our results confirm that the ancient bone specimens harbours M haplogroup signature with considerable similarities to M21ab sub-group specific to East and Southeast Asia. Sodium alginate and combinations of sodium alginate-pectin were used to study the effect on the loading efficiency and the radical scavenging activity of the polyphenols extracted from pomegranate peels (Punica granatum). The results indicate that the polyphenol content was less when the microbeads were prepared with a single type of polymer in comparison of the microbeads prepared with two types of polymers. Also there was an optimum ratio of these two polymers (2:1), which was responsible for the maximum polyphenol content. The microencapsulated particles provided to polyphenols an effective protection against the degradation phenomenon, whereas antioxidant activity remained identical. In-vitro release studies indicated that 64.87% and 48.81% of polyphenols was released in simulated gastric fluid from sodium alginate and sodium alginate-pectin microbeads respectively. 88.37% and 70.48% of polyphenols was released in simulated intestinal fluid from sodium alginate and sodium alginate-pectin microbeads respectively. An HPLC method was developed and validated to determine felodipine and related substances. A C18 column (5μm, 250 ×4.6 mm) was used for the separation at room temperature, with methanol: acetonitrile: water (50:15:35%, v/v/v) as the mobile phase at the flow rate of 1.0mLmin−1. The detection wavelength was 238 nm. The validation characteristics included accuracy, precision, specificity, linearity, and robust, and stability-indicating. The method showed good linearity for felodipine and its related substances with correlation coefficients in the range of 5.05-40.4μg/mland 0.31-15.50μg/ml. Method accuracy was assessed for felodipine and its related substances at three levels, the recovery ranged from 98.86 to 101.03%. The intermediate precision were 0.42% for felodipine and was 1.01% for the related substances (n = 9). Limit of detection and quantification for felodipine and its related substances were 1 and 4ng, respectively. The solution remains stable within eight hours at room temperature. In the present work in mathematical modeling, quantitative structure activity relationship (QSAR) studies were performed on some quinolone derivatives as a potential antibacterial agent using statistical work.
