The present investigation is mainly on the synthesis, characterization and anti-microbial screening of certain new curcuminoid analogues containing imidazole, pyrrole and thiophene rings and their metal complexes. The ability of such heterocyclic β-dicarbonyl compounds and their metal ions to influence many of complex reaction upon which the vital processes of micro-organisms depends is the motivation behind the work. A series of 5- hetero aryl-1-phenyl-4-pentene-1,3-diones(1a-c) and their Cu (II), Ni (II) complexes of ML2 stoichiometry were synthesized and characterized by UV, IR, mass and 1H NMR spectroscopies. Tinospora crispa, a plant belonging to the family of Menispermaceae is a well-known traditional medicinal plant used in Bangladesh. The methanolic extracts of stems of the plantwere evaluated for their thrombolytic and preliminary cytotoxic activities. The thrombolytic activity was evaluated using the in vitro clot lysis model while while the screening of cytotoxic activity was done using brine shrimp lethality bioassay. Different concentrations of stem extract were made in isotonic saline (2.5, 5, 10 and 20 mg/ml) and added to pre-clotted human blood (n=13) and the clot lysis activity was determined following 90 minute incubation at 37º C. The results showed that T. crispa stem extract induced mild to moderate (14.81- 25.73%) and significant (p <0.001) clot lysis activity whereas the standard Streptokinase (30,000 I.U.) evoked a significant (p < 0.001) clot lysis activity (50.1%). The present investigation highlighted the formulation and release characterization of Ketorolac Tromethamine loaded matrix tablet. Various formulations of tablets were prepared by direct compression method along with Kollidon SR and Hydroxy Propyl Methyl Cellulose (HPMC) as release retardant polymers. Each of the formulated tablets contains 50mg Ketorolac Tromethamine. The evaluation involved physical properties studies (weight variation, thickness, length, width, hardness, friability, and drug content) of tablets and in vitro release kinetics assessment. The USP paddle method was operated at 50 rpm selected to perform the dissolution test and 900 ml phosphate buffer of pH 7.4 was used as dissolution medium. The drug release from each formulation was analyzed by using release kinetics theories. All formulations followed Higuchi release kinetics. When the release data was plotted into Korsemeyer-Peppas equation, then it was confirmed that F-1 to F-5 exhibited fickian type drug release whereas F-6 exhibited non-Fickian type drug release. Solanum sisymbriifolium (Solanaceae) and Jatropha gossypifolia (Euphorbiaceae) have long been used in folkloric medicine for the treatment of various ailments. The purpose of the present study was to evaluate the cytotoxic activity of the plants and to make available the process of data analysis of cytotoxic studies to the researchers. A general toxicity of the extracts was assessed by a simple and low cost assay using brine shrimp lethality as an indicator of toxicity. Ethanol crude extract and fractions of the extract of leaves of Solanum sisymbriifolium exhibited relatively mild cytotoxic activity compared to that of flowers and fruits against brine shrimp larvae with median lethal concentration, LC50 values ranging from 10.10 – 321.35 and 13.19 – 80.16 μg/ml, respectively.
