A simple and precise stability-indicating reversed-phase (RP-HPLC) method was developed and validated for the determination of Apixaban active pharmaceutical ingredient. The chromatographic condition was carried out on Phenomenex Luna 5 µm C18 100 Å (150 × 4.6 mm) HPLC column with prepared buffer solution 1.0 ml of Orthophosphoric acid mixed with 0.2% of potassium dihydrogen phosphate solution, use as mobile phase (a), mobile phase (b) was acetonitrile. The mobile phase flow rate at 1 ml/min and the detection wavelength is 230 nm. Forced degradation study was covered acid hydrolysis, base hydrolysis, peroxide, photolytic (UV light) and thermal degradation was performed to prove the specificity of the proposed method and degradation was achieved. The aim of the present study was to improve the release rate of herbal drugs by microsponges prepared through quasi-emulsion solvent diffusion technique using ethyl cellulose and PVA as carriers. The microsponges were characterized by FTIR, DSC and SEM studies followed by the determination of total drug content and entrapment efficiency. Anti-arthritic activity was evaluated by Freund’s Complete Adjuvant (FCA) induced arthritic rat model. RA was induced by intradermal injection of 0.1 ml FCA in the left hind paw. The animals were treated with doses 50:50, 70:30, 30:70, and 100:100 mg/kg b.w. of microsponge formulation. Methotrexate at a dose of 10 mg/kg is used as a standard drug. In this present study, the objective was to extract crude saponins from the root of Phoenix pusilla (PP) and to check its inhibitory potential of α- amylase and α- glucosidase enzyme. Saponins were isolated by partitioning between butanol and water. Further, the presence of saponin was confirmed by the foam test. Then the crude saponins were analyzed by UV- spectroscopy in the range of 200-400 nm and Fourier transform infrared spectroscopy (FTIR). FTIR results revealed the presence of – OH, -C-H, C=O functional groups. In-vitro antidiabetic activity assay of isolated crude saponin with acarbose as positive standard showed efficient hypoglycemic ability. The earlier reports on Nardostachys jatamansi show contain a various class of chemical constituents that have influenced the anti-inflammatory and anti-candidal activities. The present study reports the evaluation of anti-inflammatory and anti-candidal activities of N. jatamansi and also studies pharmacokinetic properties using in-silico model. In ongoing research work, we used individual extracts of dichloromethane, ethyl acetate, methanol, and water were prepared. The anti-inflammatory activity of different solvent extracts was carried out using gelatin zymography through detection of MMP-2 and MMP-9 and on other side anti-candidal activity was carried out using Nitro blue tetrazolium assay (NBT). Bioactivity and pharmacokinetic parameters of all extracts were calculated using the property calculation online toolkit.
