The main objective of this study was to Assessment of the acute toxicity of ethanolic extract of Polyherbal formulation in Swiss albino mice. In this study, the acute oral toxicity study of ethanolic extract of PHF1 was carried out according to Organization for Economic Co-operation and Development (OECD) guideline 423 and 407 on Swiss albino mice (20–30 g). In the study, the oral dose of 2000 mg/kg of PHF1 extract was administered in the treated group. General behavior, morbidity, and mortality were determined up to 72 h and compared to control group. Further animals were investigated for a period of 3days assess any other toxic effect. Behavioral changes and other parameters such as body weight, hematological parameters, and biochemical parameters were evaluated and compared to the normal group. A simple, sensitive, and fast single-step extraction method for estimating empagliflozin in human plasma using LC-MS/MS was developed and validated for pharmacokinetics evaluations. Plasma samples were basified before solid-phase extraction on SOLA (30 mg / 1 mL cartridges). Separations were carried out on a normal reverse phase C18 column (Hypersil BDS 100 × 4.6, 5µ mm column) for 3.5 minutes at a flow rate of 0.6 mL/min. Ten µL of the SPE eluent is directly injected onto LC-MS/MS to quantify the analyte from 1.563-800.000 ng/mL using a single SRM transition (m/z: 449.140à371.100) in negative ion mode. During method validation, selectivity, matrix effect, recovery, carry-over effect, stability studies, inter-day, and intra-day precision and accuracy experiments were conducted per USFDA guidelines. Cadaba indica Lam (Indian Cadaba), an Indian traditional medicinal plant has been used for various diseases. The cold macerated methanolic leaf extract of Cadaba indica was assessed using Agilent GC 7890A gas chromatography connected with an MS- 5975C mass spectrometer and the mass spectra were matched with NIST 14.0 – data library. Several chemical constituents were identified within 28 min of the entire GC-MS analysis. The GC chromatogram shows that presence of most abundant linolenic acids and its esters such as Hexa-decanoic acid, methyl ester, n-hexadecanoic acid, 10,13-octadecadienoic acid, methyl ester, 9, 12, 15-Octadecatrienoic acid, (Z, Z)-methyl ester, Octadecanoic acid, methyl ester, 9, 12-Octadecadienoic acid, (Z, Z)-, 9, 12, 15-Octadectatrionic acid, (Z, Z, Z)-,Octadecanoic acid, Hexadecanoic acid, 2,3-dihydroxy propyl ester and 9- Octadecanoic acid, (Z) – methyl ester. Aim: The present study was to develop and evaluate antiulcer polyherbal tablet as designing of oral herbal formulation. The methanolic extracts of Abrus precatorius (MEAP) and Cordia wallichi (MECW) leaves were used in different concentrations in the formulations. Materials and Method: The current exploration was to design a gastro-retentive drug delivery system of MEAP and MECW using swelling polymer through the wet granulation method. All the formulations were evaluated for weight variation, hardness, friability, drug content, and in-vitro dissolution. In this gastro-retentive dosage form using hydroxypropyl methylcellulose-K4M was prepared to develop sustain release tablets, which could remain in the stomach for longer periods of time, delivering the drug to the site of action.
