α-amylase is widely used in various biotechnological applications such as food processing, starch, and paper industries, hydrolyzing starch, glycogen, and other polysaccharides into glucose, maltose, and oligosaccharides. Aspergillus niger α-amylase was partially purified by ammonium sulphate fractionation with 1.409 fold purity and 54.88% yield. The partially purified α-amylase was immobilized on sodium alginate with calcium chloride by ionotropic gelation with glutaraldehyde as a cross-linking agent. Immobilized α-amylase exhibited 50% of its original activity. The soluble and immobilized α-amylases exhibited maximum activity at pH values 7.5 and 8.0, respectively. The optimum temperature for both the soluble and immobilized enzymes was 35 ºC. The immobilized enzyme was more pH and thermally stable than the soluble one. The substrate starch was hydrolyzed by soluble (Km 0.6 mg/ml, Vmax 16.05 mg/ml/min) and immobilized α-amylase (Km 0.65 mg/ml, Vmax 17.41 mg/ml/min) with high efficiencies. Globally, the outbreak of antibiotic-resistant pathogens derived infections urged an intensifying necessity for the survey of underexplored marine zone microorganisms for novel metabolites. Our current study emphasized on isolation, polyphasic characterization and bioactivity of the novel actinomycetes from the Bay of Bengal near Kakinada and Nizampatnam, Andhra Pradesh, India, which could provide lead bioactive compounds of therapeutic efficacy to repress the of pervasiveness multidrug-resistant pathogens. A total of fifteen actinobacterial strains (VJRM-1 to VJRM-15) were isolated and tested. Micro-morphological characteristics of the isolates were perceived by SEM (scanning electron microscope) analysis. Phylogenetic analyses of isolates were appraised with 16S rRNA gene sequence homology. The dendrogram was contrived by deploying the parsimony method in MEGA software version 6. Based on polyphasic studies, strains VJRM-7 and VJRM-8 identified as Nocardiopsis dassonvillei (KP299158) and Nocardiopsis sp (KP299159). In-vitro antimicrobial activity of crude ethyl acetate extracts derived from these strains revealed strong antagonistic activity against Gram-positive, Gram-negative bacteria and fungi. Objective: Present research work describes an eco-friendly and quick method for the development of silver nanoparticles (AgNP) using Momordica charantia Linn. extract and silver nitrate solution. Our investigation reports the antimicrobial activity of developed nanoparticles against Staphylococcus aureus, Escherichia coli. Method: Silver nanoparticle developed by reduction of silver nitrate solution by Momordica charantia Linn. extract. Extract of Momordica charantia Linn. acts as a reducing agent. Ulta Violet Visible spectroscopy used as a confirmatory test for the development of silver nanoparticles. Result: The silver nanoparticles synthesized were confirmed by their change of color to dark brown due to the phenomenon of surface plasmon resonance. The characterization studied was done by UV-visible spectroscopy, Scanning electron microscopy (SEM), X-Ray diffraction (XRD), Fourier Transmission infrared spectroscopy (FTIR), and study antibacterial activity.
