There are many pharmaceutically important compounds from Penicillium sp. that made the fungal genera one of the most important sources for the production of bioactive metabolites. As per the results of our previous study, a bioactive metabolite, P5, from Penicillium rubens JGIPR9 had highly promising cytotoxic and anti-cancer potentials. This prompted us to undertake the current study, so as to optimize the culture conditions of this fungal isolate and thereby to enhance its growth, i.e., biomass (response 1) and production of the anti-cancer metabolite P5 (response 2). Initially, the single factor method was adopted to screen the individual culture conditions that would enhance the responses under the study. The best factors being CdCl2, glucose, amino acid concentrate, and beef extract were subjected to Central Composite Design (CCD) to identify the optimal conditions that enhance the responses. The biomass dry weight ranged from 11.9-24.1 g/L and that of P5 concentration ranged from 257-774 mg/L. The optimal conditions were verified experimentally and 3.24-fold and 12.21-fold increase were observed for responses 1...

Asenapine maleate is an atypical antipsychotic drug. It was approved by USFDA in August 2009. It is an antagonist of 5-HT dopamine and α-adrenergic receptors and a high affinity for dopamine D2 and serotonin 5-HT2A receptors. It is indicated for the treatment of various psychotic conditions like schizophrenia and bipolar disorder in adults. So it leads to the requirement of accurate and precise quantification in its bulk and buccal (effervescent) dosage form. The analytical method was developed and validated as per ICH guidelines. The proposed RS-HPLC methods fulfill the need at 100.8% accuracy with a precision of 0.25% relative standard deviation. Waters Alliance HPLC system with column Inertsil ODS 3V (150 mm × 4.6 mm, 5 µm) having UV-detector at 270 nm wavelength was used. The mobile phase having a mixture of 550 mL Acetonitrile and 450 mL of Milli-Q water and 1mL Ortho Phosphoric Acid (OPA), was used. The flow rate was set to 1.5 mL/min that gives the retention time at 4.9 min for Asenapine Maleate. The...

Spherical agglomeration technique enables simultaneous crystallization and agglomeration. Oxcarbazepine spherical agglomerates were prepared by crystallo-co agglomeration technique. Acetone acted as a good solvent, aqueous solution of PEG 6000 served as a bad solvent, and dichloromethane was used as bridging liquid. The optimization was carried out using 23 factorial design, where the factors were rotation speed, polymer-drug ratio and amount of bridging liquid. The responses evaluated were % drug release, mean yield pressure (MYP), and carr’s index. The optimized agglomerates and oxcarbazepine were evaluated by powder x-ray diffractometer (PXRD) and scanning electron microscopy (SEM). The agglomeration improved the micromeritic properties of oxcarbazepine. PXRD showed reduction in crystalline nature of oxcarbazepine, and SEM demonstrated a spherical and smooth surface. In-vitro dissolution increased by 10-fold. The AUC (35.19%), Cmax (23%), and tmax (reduced by 1h) also recorded improvement. From the results, spherical agglomerates are a suitable method and can be used to design immediate-release...

Aims and Objectives: To evaluate and compare diclofenac and meloxicam as tablets and as mucoadhesive patches in the control of odontogenic pain. Materials and Methods: 60 patients of either sex with acute dental pain were included in the study. Informed consent was obtained from each patient and they were randomly divided into 4 groups. Group A1 received meloxicam tablets, A2 meloxicam mucoadhesive patches, B1 diclofenac tablets, and group B2 was allotted diclofenac mucoadhesive patches. A 10 cm Visual Analogue Scale (VAS) was used to record pain scores at baseline, 20 and 30 min post-administration. Patients were also given the allotted tablets and patches to be used for 2 days, along with a pain diary to record their VAS scores at 30 min post-administration. Diary was collected and data analyzed on the 4th day. Results: Statistically significant reduction in pain was seen in all 4 groups from baseline to the end of the study period. Overall pain reduction was greater with the mucoadhesive patches than with tablets. Diclofenac patches showed...

The aim of the present study is to develop a stability-indicating reverse-phase high-performance liquid chromatography method for the simultaneous estimation of Aspirin and Dipyridamole in combined extended-release dosage form 25/200mg, using a single unit of the capsule. Materials and Methods: Chromatographic separation was achieved with Agilent’s high-performance liquid chromatography and X bridge C8 column, with the mobile phase containing a mixture of ammonium dihydrogen phosphate buffer (pH 2.5): methanol (550:450, v/v) by isocratic elution technique. The flow rate was maintained at 1.2 ml/min, and the detection wavelength was 230nm. Results: Aspirin and Dipyridamole were eluted at 4.7min and 12.6min, respectively, using the developed method. The method was linear in the range of 12.5-50μg/ml for Aspirin and 100-400 μg/ml for Dipyridamole, with an r2 value of 0.9997 and 0.9999, respectively. The sample recoveries observed were 97.63-100.96% and 97.38-98.01%, respectively, for aspirin and dipyridamole. The forced degradation studies were carried out, and the stressed samples were analyzed using the developed method. The purity angle of the peak was observed lesser than the...
