Objective: The present study was designed for Isolation of phytoconstituents from pharmacologically potent extracts of leaves of Cassia uniflora based on in-vitro pharmacological screening and their subsequent characterization. Methods: Crude extracts of leaves, stems, and fruits of cassia uniflora were prepared using various solvents such as water, methanol and hydro alcohol. These extracts were screened for in-vitro pharmacological activities like antioxidant, anti-inflammatory, and anti-diabetic activities. These active extracts were subjected to column chromatography by means of different mobile phases followed by TLC. The isolated compounds were subjected to IR, 1H NMR, 13C NMR, and LC-MS spectral analysis for chemical characterization. Results: The methanol extract of leaf of Cassia uniflora was found to be potent when compared with other extracts. This extract was subjected to column chromatography to get fractions and eluted fractions were run in TLC mobile phase with different solvent ratios. Delonix elata (D. elata) whole plant was chosen to evaluate in-vitro anti-oxidant activities. The total polyphenols and flavonoids were determined after the extraction. DPPH (2, 2′-diphenyl-1-picrylhydrazy), H2O2 (Hydrogen peroxide), NO (Nitric oxide) and TBARS (Thiobarbituric acid) methods were used to evaluate antioxidant activities of D. elata whole plant. The results showed that the highest levels of polyphenols were recorded in DEM (15.32 ± 0.32 mg of GA/g of extract). Total flavonoid contents of extracts/fractions showed that DEM (15.57 ± 0.69 mg of Qu/g of extract) contains the highest level. The DEM extract shows more inhibition of DPPH (92.56), NO (143.88), H2O2 (182.28) and TBARS (121.94 µg/ml), respectively when compared with other extracts. These findings suggest that the extracts obtained from D. elata stem have active substances contributing to the increase in natural antioxidant potential. Objective: To explore the Teratogenicity of isolated Polyphenols of Symphyocladia latiuscula (PSL) on pregnant female rats. Experimental Design: The marine alga Symphyocladia latiuscula were the preparation of methanolic extract followed by isolation of Polyphenols. The Teratogenic study was performed according to the OECD guidelines, no 415. And the test drug; PSL 100, 200 and 1000 mg/kg and positive control Valproic acid (VPA); 1000 mg/kg was administered from 6th to 17th day of gestation during the period of fetal organogenesis. All females were killed on the 20th day of pregnancy, and their uterus was dissected to examine the Gross Abnormalities. A simple and reliable head space Gas Chromatographic method has been developed for the determination of residual ethanol, isopropyl alcohol and toluene in Amiodarone hydrochloride drug substance. The proposed method is based on flame ionization detection technique with DB 624 as stationary phase. Linearity of detector response was established up to 150 % of the limit level for ethanol, isopropyl alcohol and toluene and the quantitation limit was 80 ppm for ethanol, 40 ppm for isopropyl alcohol and 4 ppm for toluene respectively. Performance of the method was assessed by evaluating the recovery, repeatability, reproducibility, linearity and limits of detection and quantification.
