The aim of the present study was to implement QbD approach to identify, evaluate and control critical process and formulation parameters and their impact on critical quality attributes (CQAs) and to establish design space with multifactorial combination which would provide high degree of assurance to achieve quality target product profile (QTPP). A risk assessment was performed with various process and formulation parameters to determine their impact on particle size and encapsulation efficiency (CQAs) of nanoparticles and further high risk parameters were optimized using I-Optimal design. Design space was generated by setting limits to CQAs as per acceptance criteria of Tacrolimus loaded PLGA (TC PLGA) nanoparticles. The optimized formulation from design space was further characterized by XRD, DSC, SEM, in-vitro dissolution study. The nanoparticles obtained were free flowing, spherical, smooth and uniform. In-vitro dissolution study showed ability of nanoparticles to sustain drug release for period of 4 weeks. In conclusion, it can be demonstrated that, QbD approach was successfully implemented for understanding of process and formulation parameters and further develop...

A 20-year-old male patient reported to ophthalmology outpatient department with complaint of loss of vision in right eye since childhood and diminished vision in left eye for 1month of duration. Funduscopy revealed right side optic atrophy and left sided vitreous hemorrhage. Non-contrast computed tomography study of head showed hyperdense circle of Willis and cerebral venous sinuses. No contrast was given before the computed tomography scan of head. Bilateral orbit showed right sided oval enlarged eye ball with medial deviation and normal left eye ball. Magnetic resonance venography was advised to rule out sinus venous thrombosis which revealed normal study of sinuses. On clinical examination, the patient had clubbing of all fingers, mild cyanosis, hyperemia of lower conjunctiva and systolic murmurs on auscultation with raised hematocrit level (72 percentage). Echocardiography of the patient showed features of tetralogy of Fallot (TOF). The hyper-dense vessel was due to raised hematocrit (secondary polycythemia Vera)...

Background: Snake bite is a problem across the developing countries, where agriculture is one of the main sources of income and India is one among it. Objective: The study was carried out to determine the pattern of snake bite in four different selected tertiary care hospitals of south India for a period of 03 years from March 2011 to March 2014. Material and Methods: A prospective observational study of 270 patients of snake bite was collected in patients admitted to emergency department in four different selected tertiary care hospitals in south India. Patients with history of previous visit for treatment before admitting into the study sites were excluded from the study. Results: There were 201 males [74.4%] and 69 females [25.5%] out of 270 snakebite patients in our study. Of that, 142 [52.6%] patients were between the age group of 30-50 years. Most of the cases were farmers [56.66%] and Laborers [32.96%]. Peak incidence of snake bite was observed during rainy season [68.8%]. Most of the envenomation had reported...

Addicted smokers Semen analysis from a cross section of population of Anantapuramu district, Andhra Pradesh, India was done as it provides a useful profile of the function of the male reproductive health. Seventy two (72) men were drown from a cross section of smokers and divided in to two age groups namely 25-30 years and 31-40 years. In both age groups persons were selected basing as how many cigarettes they smoked in a day. Different semen parameters like volume, liquefaction, alkalinity, sperm count, motility and morphology were analysed. Incidences of Hypospermia in 25, Teratozoospermia in 27, Oligozoospermia in 54, Oligoasthenozoospermia in 20, and Oligoasthenoteratozoopermia in 6 persons were observed. Abnormalities were seen in both age groups and in persons who smoke either moderately or...

In Present study the antinociceptive and the gastroprotective effects of orally administered or inhaled Thevetia peruviana Pers. K. (Schum) (Oleander) volatile oil, and its principal constituents linalool and 1,8-cineole were evaluated in mice. Either when orally administered (100µl/kg) or inhaled for 60 min Thevetia peruviana volatile oil significantly reduced the acetic acid-writhing response in a naloxone-sensitive manner. In the hot plate test, analgesic activity observed after oil inhalation was inhibited by naloxone and atropine sulphate pretreatment suggesting the involvement of opioidergic as well as cholinergic pathways. Regardless of the administration route and the experimental model used both linalool and 1,8-cineole did not produce significant analgesic response. Oral or inhalatory treatment with analgesic doses of volatile oil did not affect mice spontaneous locomotors activity. Concerning the gastric effects, Oleander oil, linalool and 1,8-cineole oral administration protected against acute ethanol-induced gastric ulcers but did not prevent indomethacin-induced lesions indicating no interference with arachidonic acid metabolic cascade. In conclusion, besides this gastroprotection, Oleander oil reveals an interesting analgesic activity mainly relevant after...
