The aim of the present study was to develop and validate a new, simple, selective and economical stability indicating RP-HPLC method for the quantitative determination of resveratrol in bulk drug and pharmaceutical formulations. A Box-Behnken design supported optimization was carried out to identify the optimum chromatographic conditions. The developed method was validated for linearity, range, accuracy, precision, reproducibility, LOD, LOQ and robustness as per ICH guidelines. Forced degradation studies were carried out in different stress conditions to detect degradation peak using validated method. Optimum chromatographic separation was achieved by mobile phase consisting of methanol, water and acetic acid in 69:30:1 ratio respectively. The flow rate of 1 ml min-1 with standard RT of 2.8 min was optimized in the present study. The method was linear in the concentration range of 7.5-60 µg mL-1 with a regression coefficient (R2) of 0.999. The LOD and LOQ was found to be 1.463 and 4.737 µg mL-1 respectively. Aim: The aim of the study is to assess effect of lifestyle modifications and implementation of DASH diet in daily life on blood pressure in hypertensive patients. Method: A prospective observational study was conducted for a period of six months from Dec- 2013 to May-2014. The data was collected by self prepared form and analyzed using descriptive statistics. Counselling on lifestyle changes and implementation of The DASH diet were given to the study population and post counseling changes were collected at visit 2. The significant difference in systolic and diastolic blood pressure was calculated by unpaired t-test. Results: A total of 186 patients were enrolled in the study, the mean (SD) age was 55.89(9.44), the percentage distribution of the study population showed that 103(55.37%) females and males 83(44.62%). A simple, precise, and robust spectrophotometric method for the determination of Ceftriaxone sodium has been investigated and validated. The proposed chemical reaction involved involves the reaction primary amine of Ceftriaxone sodium with nitrite solution in acidic medium to form a diazotized product followed by the reaction of the diazotized product with suitable coupling reagent to obtained a colored product. Ceftriaxone sodium has shown λmax 510nm with absorbance of 0.126. The method obeys Beer Lambert law at concentration range of 1-20ppm by showing linearity curve between absorbance and concentration. The objective of present study was to develop effervescent tablets of promethazine (PMZ) for the treatment of emesis. Effervescent tablets were prepared by direct compression method and were optimized using 32 full factorial design. Amount to sodium starch glycolate(X1) and amount of sodium bicarbonate (X2) were selected as independent variables, whereas disintegration time (Y1), amount of carbon dioxide (Y2) and drug release in 5 minutes (Y3) were selected as dependent variables. All the batches were also evaluated for general post compression evaluation of tablet such as- weight variation, thickness, friability and hardness. From the results of design batches, best batch was selected and evaluated for in vivo pharmacokinetic study in a rabbit model. The disintegration time ranged from 58.67±0.27sec to 228.67± 0.67sec while amount of carbon dioxide ranged from 0.1±0.082 gm to 0.29±0.061gm in all the design batches. Neuropathic pain is a persistent chronic pain caused by primary lesion or dysfunction of neurons which is primarily characterized by hyperalgesia, allodynia. There is some evidence that acamprosate might inhibit Ca2 influx through NMDA and voltage-dependent Ca2 channels. λ-carrageenan (0.1 ml of 1% w/v) into i.pl. region of the hind paw was injected to produce acute inflammation. Unilateral peripheral mononeuropathy was induced by sciatic nerve ligation. Thermal and mechanical hyperalgesia and cold allodynia were assessed. The extent of oxido-nitrosative stress was assessed by estimating the levels of thiobarbituric acid reactive substances, catalase and superoxide dismutase activity and nitrite levels in both sciatic nerve and dorso-lumbar spinal cord homogenate in CCI model.
