A Simple, rapid, sensitive, accurate and precise spectrofluorimetric method was developed for the determination of tolterodine tartrate (TOL) in bulk powder and in pharmaceutical preparation. This method is based on measuring the native fluorescence of TOL in methanol at 313 nm after excitation at 285 nm using 1 ml of phosphate buffer pH 6. All variables that affect fluorescence intensity such as diluting solvents, pH, buffer and buffer volume were studied and optimized .The fluorescence-concentration plot was rectilinear over the range of 0.5-8 µg/ml with a lower detection limit (LOD) of 0.090 µg/ml and lower quantification limit (LOQ) of 0.273 µg/ml. The method was validated and successfully applied to the determination of TOL tablets with an average percent recovery ± RSD% of 100.57 ± 0.76. The obtained results were statistically compared with those of the reported method by applying t-test and F-test at 95% confidence level and no significant difference was observed regarding accuracy and...

A simple, rapid and validated high performance liquid chromatography (HPLC) method with UV detection for the quantification of atorvastatin Calcium microemulsions (ATC-MEs) in rat plasma was developed. The assay procedure involved chromatographic separation using a C-18 chromatographic column (250 mm × 4.6 mm, i.d., 5 μm) and a mobile phase of methanol/water (containing 0.05 % glacial acetic acid) (70:30, v/v) 1.0 mL/min and detection wavelength of 248 nm. Plasma sample (200 μL) pretreatment was based on simple deproteinization by ethanol and centrifugation. The organic layer was evaporated under N2 gas, reconstituted with 200 μL of mobile phase, and 20 μL portions of reconstituted sample were injected onto the column. Calibration curve was linear (r ≥ 0.9854) from 0.1 to 50 mg/L. Both intra- and inter-day assay precisions that are presented through RSD were lower than 12.4% for intra-day and lower than 12.5% for inter-day assessment. This method was omitting the use of expensive solid phase extraction and time consuming liquid extraction procedures. Moreover, the present method was successfully applied...

Pelletization can be defined as an agglomeration process that converts fine powders or particles of a bulk drug and excipients into small, free-flowing, more or less spherical units, called pellets. The size of the pellets is 0.5-2mm. Pellets have the free flowing capacity and have low porosity about 10%. Preparation methods include direct pelletizing, powder layering,Suspension or Solution layering, pelletization by extrusion and spheronization, spherical agglomeration, compression/balling,Cryo pelletization, melt spheronization, globulation or droplet formation, fluid bed coating. Pellets have various advantages when compare to normal conventional dosage forms, like they help in giving accurate dosage to the paediatrics and geriatrics and even to the bed ridden persons, reduces peak plasma fluctuation, Minimize potential side effects without lowering bioavailability, avoiding high local concentration, Less susceptible dose dumping. At present usage of pellets has increased largely due to their advantages and there novel approaches, the novel approaches of pellets includes; 1). They help in preparation of modified release multiple dosage form with different release patterns like immediate and sustained release pattern, 2) They help...

Arthritis is still a challenge for medical research. Pharmaceutical research has resulted in several new approaches for treatment/management of arthritis including drugs like the biologic disease modifying anti-rheumatic drugs (DMARDs). Several disadvantages like serious side effects, high costs and requirement of parenteral administration still invite more research in this area to provide a convenient, affordable therapy with lesser or no side effects. Traditionally used herbal medicines, due to their anti-inflammatory and immunomodulatory properties, have potential to be a therapy of choice for arthritis patients and are now extensively being studied. Although a number of these medicines are being used traditionally for their therapeutic activity, development of their novel drug delivery systems was not attractive to the scientists due to insufficient knowledge about their exact mechanism of action and difficulties in processing, standardising, extracting and identification of active constituents. Current research demonstrates that the effectiveness and bioavailability of herbal actives can be improved by understanding of exact mechanism of action and by formulating them into novel technologies like liposomes, phytosomes...
