The present work deals with development and standardization of phytochemical screening for quantification of methanolic extract of medicinal plant stem bark of Anthocephalus cadamba (Roxb.)Miq. The scientific parameter is necessary to identify the exact plant material and to find its quality and purity. The present study deals with various, physical evaluation and preliminary phytochemical screening of various successive extracts such as qualitative chemical analysis and different spectroscopic study were carried out and the parameters were reported. HPTLC of extract shows the seven different peaks confirming that the seven compounds present in methanolic extract of Anthocephalus cadamba. A novel method was developed for the determination of fluoroquinolones such as ciprofloxacin hydrochloride (CPL) and ofloxacin (OFX) by isocratic reverse phase-high performance liquid chromatography (RP-HPLC) coupled with UV detection. The developed method was rapid, accurate, reproducible, economical and sensitive for the simultaneous estimation of ciprofloxacin hydrochloride and ofloxacin. Fluoroquinolone antibiotics were separated on an analytical column (SIL 100Å, 125×4.6 mm ID) C18-RP, 5 µm, at ambient temperature. The mobile phase was consisted of Phosphate Buffer (15 mM): Methanol: Acetonitrile: TEA (66:24:10:1% v/v), pH was 6.5 (adjusted with 80% HCl), at a flow rate 1ml/min and sample injection volume was 20-µL. UV detection at 289 nm and retention time (RT) was 3.01 and 6.03min for ciprofloxacin hydrochloride and ofloxacin respectively. The method was validated in terms of stabilizing accuracy, precision (Intra/Inter-day), linearity, specificity, stability and sensitivity. The effect of Adenanthera pavonina (AP) seed extract on the blood cholesterol level of atherogenic diet rats was evaluated. Twelve adult male Wistar rats divided into 3 groups of 4 animals each were used and were treated orally with normal saline (Control group), atherogenic diet (Positive control) and 200mg/kg of AP seed extract (treatment) over a 4-week period. The present study was undertaken to assess body weight, total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL) and very low density lipoprotein (VLDL). They were significantly lower in the extract treated groups compared to the control group. Phytochemical screening showed that the extract contained cardiac glycosides, tannins, saponins, alkaloids and flavonoids. Cyanogentic glycosides and anthraquinones were absent. Histopathological examination showed that the extract did not cause any significant lesion changes in the liver, kidney and even the testes.
