Paracetamol, caffeine, phenylephrine, and chlorphenamine maleate are available on the market to treat pains, psychoactive disorders, and bronchopulmonary disorders. The objective was to develop a single HPLC method to determine paracetamol, caffeine, phenylephrine HCl, chlorphenamine maleate, and its impurities in flu cold tablets combination dosage form. No analytical method is available to assess the quality of flu cold tablets’ combination dosage form; hence the present research was taken. The evaluated method was validated with Inert sustain C18 AQ 250 mm x4.6 mm, 3µm column, flow rate of 0.7 mL/min, 220 nm wavelength, 50 µL injection volume, 35 °C column temperature and gradient program run time 160 min and also used the pH 2.5 and 4.0 phosphate buffer. This single method was developed and validated with precision, accuracy, ruggedness, linearity, robustness, and specificity by following ICH and USP validation of compendial procedures. The degradation study was performed at all stress conditions such as water hydrolysis, acid hydrolysis, base hydrolysis, oxidation, thermal and light exposure, the peak purity of each active...

Dental caries is one of the serious bacterial diseases of the oral cavity caused by diverse Streptococcus species. Streptococcus mutans strains displayed remarkable genotypic and phenotypic diversity. Glucosyl transferase genes are recognized as essential enzymes leading to biofilm production, the destruction of the ecosystem of normal bacterial communities, and the development of dental caries. The purpose of this study is to detect S. mutans associated with Dental Caries among adults. In our studies, a total of 50 plaque specimens, 23 from male and 27 from female from dental caries patient’s samples, were collected from dental clinics in Jeddah province. All bacterial isolates were identified by culture on Tryptone Yeast Extract Cystine w/Sucrose (TYCSB), Bile esculin slant agar, and Blood agar (BA) media. The results of the current study revealed that only four (8.0%) bacterial isolates out of 50 plaque specimens were identified as S. mutans. These four bacterial isolates named RHA1, RHA2, RHA3, and RHA4 were identified by biochemical and molecular tests. Partial DNA fragment of Glucosyl transferase (GtfB)...

The purpose of the present investigation is to evaluate the applicability of hupu gum and lannea gum in the design of buccoadhesive patches for extended-release over a period of 6 hours, using Salbutamol sulphate as a model drug. The efficiency of these polymers was compared with already established synthetic polymer PEO coagulant. The patches were prepared by using the solvent casting method in which, initially backing layer with cellulose acetate was formed on which patches were cast for unidirectional drug release to buccal mucosa. In-vitro drug diffusion studies were done by using Franz diffusion cells. The kinetics of both the phases were calculated, and both the phases followed zero-order for the majority of the patches. Based on the swelling Index (%), buccoadhesive strength, ex vivo residence time and in vitro diffusion studies SHG9 (hupu gum), SLG9 (lannea gum) and SPEO6 were considered as optimized formulations as they could extend the drug release over a period of 6 hours as contemplated. Among three optimized patches, SHG9 and SLG9 were found...

Indigenous plants like Withania coagulans fruits. Alstonia scholaris bark and Aegle marmelose leaves have been used for diabetes management by traditional Indian people for a long time. The present study was carried out to investigate anti-hyperglycemic potential of an herbal mixture comprising of extracts of these three plants in streptozotocin-induced diabetes in Wistar rats and to focus on its possible mechanism of action. Experimental diabetes was induced in Wistar rats by a single intraperitoneal injection of streptozotocin (65 mg/kg). Animals were divided in five groups (n=6) and treated with herbal mixture (100 & 200 mg/kg) for 4 weeks. At the end of the study period, fasting blood glucose, oral glucose tolerance, glycol-sylated hemoglobin, serum insulin, glucose uptake from rat hemidiaphragms, and liver glycogen were determined. Antioxidant enzymes of the liver, including superoxide dismutase and catalase, were evaluated. Histopathology of the pancreas was studied. The results of our study demonstrate anti-hyper-glycemic and antioxidant potential of the herbal mixture, justifying its use in the indigenous system of...

Background: The present work aimed to evaluate the sustained release antipsychotic activity of formulated biodegradable mucoadhesive guar gum microspheres of atypical antipsychotic drug Olanzapine (OLZ) for intranasal delivery and to reduce the dose-related side effects of the drug olanzapine. Guar gum microspheres of Olanzapine were fabricated and designed by O/W emulsion solvent evaporation technique using non-halogenated solvents and PVA and tween-80 as surfactant and stabilizer. The prepared guar gum microspheres were characterized for various physical and micrometric properties. Box Behnken design of experiment was applied for the formulation of various batches. An optimized batch was selected for in-vivo studies. Objective: This study solely emphasizes on sustained effect of formulated Olanzapine microsphere on different symptoms of schizophrenia. Material and Methods: Rats of Charles foster strain were subjected to a battery of various behavioral tests for representing positive, negative, and cognitive impairment symptoms found in schizophrenic patients. Different tests performed were open field exploratory, elevated plus maze, forced swim, passive avoidance and Morris water maze. Results and Conclusion: During in-vivo studies,...
