The objective of the present investigation was to formulate the thermosensitive ophthalmic in-situ gels of Dorzolamide HCl for the treatment of glaucoma. Dorzolamide HCl is presently available as conventional eye drops, which has demerits like pre-corneal elimination, lachrymal drainage and no sustained effect leading to poor bioavailability and hence there was a need to develop a better, stable and sustained drug release dosage form. Dorzolamide HCl loaded thermosensitive ophthalmic in-situ gels were prepared by cold method using poloxamers as thermo-sensitive polymers in different concentrations with HPMC K4M as viscosifier which was used in three different concentrations. A total of six formulations were made and evaluated for para-meters, namely appearance, clarity, pH, gelling capacity, gelation temperature, rheological study, drug content, and in-vitro drug release studies. The medicinal plants, Adhatoda vasica, and Adhatoda beddomei are commonly used in the treatment of several illnesses and respiratory disorders viz., cold, severe cough, chronic bronchitis, asthma and tuberculosis. The present study aimed to identify the phytochemical constituents and quantification of bioactive contents of hexane extracts of these plants, followed by metabolic fingerprinting, which was carried out by GC-MS analysis. Assessment of phytochemical screening showed the presence of various phytochemicals like glycosides, phytosterols, flavonoids, terpenoids, and phenols in hexane extract of A. vasica and A. beddomei. A rapid method has been used for the comparative study of A. vasica and its related plant species A. beddomei using gas chromato-graphy-mass spectrometry (GC-MS). The phytocompounds comparison in two plants by GC-MS was similar in 12 compositions. The objective of the present study is to evaluate the phytochemical constituents, antioxidant and antibacterial activities of methanolic leaf extract of Clinopodium nepeta (L.) Kuntze. Total phenolic content of leaf extracts was assessed using Folin-Ciocalteu and AlCl3 reagent. Antioxidant assays were evaluated using DPPH, ABTS radical scavenging, and ferric, reducing antioxidant power (FRAP) methods. The leaf extract was screened against four pathogenic bacteria, two gram-positive (Escherichia coli, Bacillus cereus) and two-gram negative (Pseudomonas aeruginosa, Klebsiellea pneumonia) using disc diffusion method. The results showed the total phenolic content (378.52mg gallic acid equivalent (GAE)/g) and flavonoid content (189.4 mg quercetin equivalents (QE)/g) of methanolic leaf extracts. DPPH and ABTS radical scavenging activity with an IC50 value of 0.49mg/ml and 1.68mg/ml, respectively. It also exhibited good absorbance in the FRAP assay. The in-vitro antibacterial assay showed an excellent zone of inhibition against Pseudomonas aeruginosa (19.01±0.3mm), Bacillus cereus (24.66±0.3mm), Klebsiellea pneumoniae (16.74±0.3mm), and Escherichia coli (17.56±0.31). The aim of this study was to prepare a suitable mucoadhesive in-situ gel formulation of clotrimazole for the treatment of vaginal candidiasis. The in-situ gel-forming systems are viscous liquids that turn to a gel phase upon exposure to physiological conditions. The lower retention of conventional formulations in the vagina, mucoadhesive formulations would prolong the retention time. For this study mixture of poloxamer (Plx) 407 and 188 were used. 32 full factorial design was applied for the optimization.
