An analytical method consists of a detailed, stepwise list of instructions to be followed in the qualitative, quantitative, or structural analysis of a sample for one or more analytes and using a specified technique. A novel, simple, precise, sensitive, and reproducible RP-HPLC method for the simultaneous estimation of Dapagliflozin and Saxagliptin in bulk and pharmaceutical formulation was developed and validated. The separation was carried out on Symmetry C8 (4.6 × 150 mm, 3.5 µm, Make: XTerra) column with buffer: acetonitrile in the ratio of 70:30 %v/v (pH 3) as the mobile phase at the flow rate of 1 ml/min. The eluent detection was carried out using a UV-Visible detector at 221 nm. The retention time of Dapagliflozin and Saxagliptin was 2.83 min and 4.35 min, respectively. Linearity was observed Dapagliflozin and Saxagliptin in the concentration range of 25-125 μg/ml and 12.5-62.5 μg/ml, respectively. Calotropis procera (Ait) R. Br. (Asclepiadaceae) is a small, hardy, pubescent, evergreen, erect and compact shrub that grows wild in South-Eastern Asia. It is prescribed to treat anasarca, asthma, ascites, bronchitis, cough, cutaneous diseases, intestinal worms, leprosy, and eczema. A methanolic extract of the leaves of Calotropis procera was subjected to chromatography over silica gel column to isolate as six phytoconstituents, and their structures were characterized as n-nonanyl octadec-9-enoate (n-nonanyl oleate, 1), lup-12, 20 (29)-dien-3β-olyl hexadecanoate (lupenyl palmitate, 2), 1-methyl-4-(2′-n-decanyl)-cyclohex-1-en-5-one (3), (Z)-eicos-9-enoic acid (gadoleic acid, 4), n-decanoyl-α-L-arabinopyranoside (n-capryl arabinoside, 5) and stigmast-5- en- 3β- ol- 3-O-β-D-glucopyranosyl-(1→2)-β-D-glucopyranoside-2′′-hexadecanoate (β-sitosterol-diglucosyl palmitate, 6). A simple, precise, accurate, sensitive and robust stability-indicating HPLC method for simultaneous estimation of propylthiouracil and its impurity thiourea has been proposed. The separation was achieved on a C18 column (4.6 mm × 150 mm, particle size 5.0 μm) maintained at 45 ºC with a mobile phase composed of water: methanol: acetonitrile (50:35:15 v/v/v) with 0.1% acetic acid and detection wavelength was 241 nm. In statistical analysis, the linear response in the range of 30 – 300 μg/ml for propylthiouracil and 0.3 – 30 μg/ml for thiourea with a correlation coefficient greater than 0.99 was obtained. In forced degradation studies, PTU was found to degrade under basic hydrolysis, oxidative and photo stress while found resistant to acid/neutral hydrolysis and thermal degradation. The probable structures of six major degradants generated under stress conditions were identified by LC-MS studies and the most likely degradation pathway was proposed from mass spectral data. Introduction: Nymphaea pubescens Willd. is one of the well-known medicinal plants, being used from ancient times in India and commonly called “Kumuda”. It is an aquatic perennial herb. It has leaves having a smooth upper side, a strong root, and a branched rhizome. According to Bentham and Hooker classification “Genera Plantarum”, this plant belongs to the division Polypetale of Dicotyledones, which has been divided into 15 order and 84 families. Nymphaea comes under the Araceae family, which itself contains 8 genera and 100 species. Method: Morphological characters have been examined through a transverse section cut by microtome.
