Trigonella foenum-graecum Linn. is used in folk medicine as alcoholic and aqueous preparations for prevention of pregnancy. Pregnane- progesterone is one of the important steroid has direct connection with contraceptive activity. Pregnane is a C21 steroid and indirectly, a parent of progesterone. The present work was carried out to find out the responsible phytochemical for contraceptive activity. The objective of this study was to provide an alternative contraception and thus evaluate the pregnane progesterone. Quorum-sensing is a phenomenon in which bacteria exhibit several density dependent phenotypes. Quorum-sensing is achieved through production of auto – inducer which on acquiring a threshold concentration, activates the genes, bringing into effect the concerned phenotypes. Ayurvedic medicinal plant extracts are being used to treat a plethora of infectious disease and are known to be rich in phytochemical compounds. This work aims at screening Rosemarinus officinalis and Valeriana officinalis for their anti- quorum-sensing activity using microbial biosensor strains which includes Pseudomonas chlororaphis (aureofaciens) 30-84, S. aureus agr P3::blaZ pRN8826 and C. albicans 10231. Initially, the medicinal plant extracts are made and minimum inhibitory concentration (MIC) of these extracts is determined. Human adipose tissue-derived stem cells (hADSCs) are promising for cellular therapy as they are multipotent, which consistently express high CD90, CD105, CD73 and low hematopoietic surface markers. Flow cytometry is a routine technology that is used in most laboratories and can be used to study hADSC characterization, but the instrument needs strict calibration to assure the quality of the results. However, for rarely used fluorophores, calibration is often neglected. This study aimed to compare calibrated and non-calibrated fluorescence (FL) in flow cytometry performances. hADSCs were isolated from lipoaspirate and cultured in human platelet lysate supplemented medium. Passaged cells were used for characterization. Cells stained with positive (CD90-FITC, CD105-PerCP Cy5.5, CD73-APC) and negative (CD34, CD45, CD11b, CD19, HLA-DR)-PE marker cocktails were subjected to flow cytometry analysis. Anti-inflammatory activity of the SC-EA extract of the marine Streptomyces carpaticus was studied in Wistar rats using acute and chronic inflammatory models like carrageenan induced paw edema and cotton pellet induced granuloma models respectively. The acute toxicity of SC-EA extract was tested on adult Wistar rats. The maximum dose of SC-EA extract at 2000mg/kg does not show any sign of visual changes in physical behaviour, protein level and enzyme activities of AST, ALT, and ALP after 14 days of administration in rats. The SC-EA extract of 50mg/kg and100 mg/kg, p.o. demonstrated the inhibition of paw edema to 42.18% and 47.08% respectively and standard diclofenac showed 46.78% inhibition after 4 h of carrageenan treatment. In cotton pellet induced granuloma, maximum inhibition was accounted with SC-EA extract 58.3% at a dose of 100mg/kg and the suppression of inflammation was comparable to Diclofenac (30mg/kg), which reduced the weight of cotton pellet granuloma by 56.5%. Justicia wynaadensis, an endemic medicinal plant found in the regions of Western Ghats is a herb, belonging to the family Acanthaceae. The following study was carried out to evaluate the in vitro cytotoxicity of the leaf extract of Justicia wynaadensis. Four different extracts were prepared by using water and methanol [aqueous fresh leaves extract, methanolic extract of dry leaves powder, hot (boiled) and cold (macerated) aqueous extracts of dry leaves powder]. Preliminary screening of extracts showed the presence of flavonoids, phenols and terpenoids and its quantitative estimation was done using HPLC. Antioxidant activity was evaluated by DPPH and ABTS assay. The cytotoxic effect of all the four extracts were tested by MTT assay on two cancer cell lines MCF7 (Breast cancer cell line) and HCT116 (Colorectal cancer cell line).
