One of the earliest traditional medical systems, Ayurveda, has become well-known around the world for its extensive use of herbal formulations and holistic treatment approach. Concerns about quality, safety, effectiveness, repeatability, and standardisation continue to be significant obstacles despite the growing commercialisation of Ayurvedic goods. Therefore, in order to prove the legitimacy of commercialised Ayurvedic medicines in contemporary healthcare systems, scientific validation has become crucial. Pharmacognostic evaluation, phytochemical screening, chromatographic profiling, physicochemical analysis, toxicity assessment, pharmacological investigations, and clinical validation are among the methods now employed in the scientific validation of Ayurvedic formulations that are included in this study. The paper also emphasises quality control methods, contemporary analytical techniques, regulatory frameworks, and difficulties in standardising polyherbal formulations. Integration of traditional Ayurvedic principles with modern scientific methodologies is necessary to ensure safety, efficacy, consistency, and global acceptance of Ayurvedic...

Obesity is a chronic, complex disorder with a rapidly increasing global incidence, affecting both children and adults and significantly contributing to morbidity, mortality, and healthcare expenditures. It is strongly associated with noncommunicable diseases like type 2 diabetes, cardiovascular disease, and other cancers. Obesity’s pathophysiology is complex, including appetite dysregulation, energy balance, and metabolic pathways, with emerging evidence pointing to the involvement of brain insulin resistance and altered hormonal signaling. In cretin hormones, notably glucagon-like peptide-1 (GLP-1) and glucose-dependent insulin tropic polypeptide (GIP), control glucose metabolism and satiety. As a result, GLP-1 receptor agonists and dual incretin-based drugs have developed as effective weight-loss treatments. Semaglutide, liraglutide, and tirzepatide have all shown considerable weight loss and improvement in cardiometabolic indicators in both clinical studies and real-world settings. Among these, semaglutide and tirzepatide are quite successful, causing significant and long-term weight loss. These medicines primarily function by reducing appetite, slowing stomach emptying, boosting insulin secretion, and regulating central nerves system (CNS) pathways involved with hunger and reward. Furthermore, they have a favorable...

Schizophrenia, a chronic neuropsychiatric disorder, requires long-term administration of antipsychotic drugs, many of which suffer from poor aqueous solubility and low oral bioavailability. These pharmacokinetic limitations often result in subtherapeutic exposure, variable patient responses, and an increased risk of non-compliance. Nanosuspension-based drug delivery has emerged as a promising strategy to overcome these challenges by enhancing the dissolution rate and systemic absorption of poorly water-soluble antipsychotics. This review explores recent advancements in nanosuspension technology, including top-down and bottom-up fabrication methods, surface modification strategies, and targeted delivery approaches. Emphasis is placed on the role of nanosuspensions in optimising the pharmacokinetic profiles of atypical antipsychotics such as risperidone, olanzapine, and quetiapine. Furthermore, we highlight preclinical and clinical data supporting their use, discuss regulatory considerations, and propose future directions for personalised nanosuspension-based schizophrenia therapy. Overall, nanosuspension formulations offer a transformative avenue for improving drug bioavailability, therapeutic efficacy, and patient adherence in schizophrenia...

Polycystic Ovary Syndrome (PCOS) is a prevalent endocrine disorder and a leading cause of anovulatory infertility in women of reproductive age. Conventional oral therapies, such as clomiphene citrate, are often limited by poor bioavailability, first-pass metabolism, systemic side effects, and reduced patient compliance. Recent advancements in nanotechnology have introduced solid lipid nanoparticles (SLNs) as promising drug carriers due to their biocompatibility, stability, and controlled drug release properties. This review focuses on the application of SLN-incorporated transdermal drug delivery systems (TDDS) for PCOS management. Transdermal patches offer advantages such as bypassing hepatic first-pass metabolism, sustained drug release, and improved patient compliance. The review also highlights skin structure, formulation strategies, and evaluation parameters relevant to TDDS. Overall, combining SLNs with transdermal patches presents a novel and effective approach to enhance therapeutic outcomes and minimize adverse effects in the treatment of...

Objective: The present study aimed to develop and validate a simple, rapid and environmentally acceptable HPTLC method for the quantitative estimation of bupropion hydrochloride in tablet dosage form and to evaluate its greenness profile using AGREE, GAPI, NEMI and Analytical Eco-scale tools. Methods: Chromatographic separation was achieved on silica gel 60 F254 precoated aluminium plates using methanol: chloroform: glacial acetic acid (1.5:8.5:0.3, v/v/v) as the mobile phase. Densitometric analysis was performed at 252 nm after a chamber saturation time of 15 min. The method was validated according to ICH Q2(R1) guidelines. Results: Bupropion hydrochloride exhibited a compact spot with an Rf value of 0.37 ± 0.001. The method showed linearity over the concentration range of 150–350 ng/band with a correlation coefficient of 0.9984. The assay of marketed tablets was found to be 98.44%. The recovery range was –%, indicating good accuracy. Precision studies showed %RSD values below 2%, confirming method precision. The LOD and LOQ were found to be 7.36 ng/band and 22.31 ng/band, respectively. Robustness studies indicated that...
