A new, simple, accurate, linear, precise, efficient and reproducible Reverse Phase-High Performance Liquid Chromatography (RP-HPLC) method for the simultaneous determination of Ambroxol and Doxofylline in combined tablet dosage form. The chromatographic method was standardized using a Kromosil C18, 150mm × 4.6 mm, 3.5 ? (particle size), Thermo scientific from Germany with isocratic conditions, and mobile phase containing potassium dihydrogen orthophosphate buffer-pH 6.8 (0.01M KH2PO4): acetonitrile (25: 75) at flow rate of 1ml/min using UV detection at 257 nm. The retention times of Ambroxol and Doxofylline were 5min and 2min, respectively. The method was linear over the concentration range for Ambroxol 0.6–36 ?g/mL and for Doxofylline 16-96 ?g/mL. The recovery of Ambroxol and Doxofylline was found to be in the range of 98.13–99.85% and 98.09–99.66%, respectively. The validation of method was carried out using ICH guidelines. Salacia reticulata, used in traditional and local medicines in Western Ghats of Southern India for the treatment of hyperglycaemia, is widely used for its sugar lowering activity. Accordingly, the present study was designed to investigate the possible actions of ethanolic extract of roots of Salacia reticulate (EESR) and an ethanolic extract of local formulation (EELF) made by mixing roots of Salacia reticulata along with five other drugs, each one in equal ratios, namely heartwood of Pterocarpus marsupium, stem of Tinospora cordifolia, fruits of Phyllanthus emblica, roots of Vetiveria zizanioides, and heartwood of Acacia catechu, on sugar lowering effect of diabetic rats. In the first set of experiments, hypoglycaemic effects of oral administration at various doses (50 and 100mg/kg) of the both extracts (EESR & EELF) were examined in normoglycaemics, glucose-induced hyperglycaemic rats. Emergence of antibiotic resistance has become a serious problem worldwide. The extensive and uncontrolled use of antibiotics increases the number of multi drug resistant (MDR) bacterial strains. The major mechanisms that bacteria evolve to develop resistance against β-lactam class of antibiotics are the production of β-lactamases and expression of low affinity penicillin binding proteins (PBPs). These PBPs are involved in the final stages of peptidoglycan synthesis. Guillain-Barré syndrome (GBS) is an acute inflammatory, autoimmune disorder of peripheral nervous system. Association of TLR2 and IL-8 polymorphism with higher expression level has already been studied in many inflammatory and autoimmune diseases. However, the possible role of TLR2 and IL-8 polymorphism in GBS remains unknown. Therefore, the current study investigated the TLR-2 (Arg677Trp & Arg753Gln) and IL-8 (-251A/T) polymorphisms in 105 GBS patients and 100 healthy controls using polymerase chain reaction-restriction fragment length polymorphism analysis. Further, the expression of TLR-2 and IL-8 gene was determined by western blotting and enzyme-linked immunosorbent assay respectively. TLR2 (Arg677Trp & Arg753Gln) heterozygous genotypes were strongly associated with increased risk of GBS. TLR2 and IL-8 genes showed significantly higher expression in GBS when compared with healthy controls. There are various studies on growth of Streptococcus mutans which was done by different scientists on different media but it has been found that there is still some deficiency in existing medium. In our study we have tried to prepare a modified, less laborious and more selective medium for Streptococcus mutans which is specific for their growth. We studied 10 plaque samples from caries active school children and 2 reference strains of Streptococcus mutans ATCC 25175 (serotype c) and OMZ 61 (serotype a) to evaluate the selectivity and sensitivity of our modified medium i.e TSBB (Thioglycolate Sucrose Blood Bacitracin) agar. To check efficiency of our modified TSBB medium, both reference strains and plaque samples were plated on TSBB and other four old existing media i.e TYCSB, TSY20B, MM10, and GSTB for their comparison and evaluation.
