The oral route is considered to be the most preferred route for administration of drugs for systemic effect, but the oral route is not suitable to the administration of drug for lower gastrointestinal (GI) diseases, this happened due to their release at upper GI tract (stomach, small intestine), which further minimizes the accessibility of drugs at the lower GI tract. To overcome this difficulty, colon-specific drug delivery systems have been broadly analyzed during the last two decades. Colonic drug delivery has gained increased importance not just for the delivery of the drugs for the treatment of local diseases associated with the colon like Crohn’s disease, ulcerative colitis, etc. but also for the systemic delivery of proteins, therapeutic peptides, anti-asthmatic drugs, antihypertensive drugs, and anti-diabetic agents. This review article discusses, in brief, the introduction of the colon, factor affecting the colonic transition, colonic diseases and the novel and emerging technologies for colon targeting....

Saponin derivatives from Vietnamese ginseng are proven for their efficacies in modulating oxidative stress, but there had been no reports about the interaction between them and two enzymes, endothelial nitric oxide synthase (eNOS) and cytochrome P450 2E1 (CYP2E1). eNOS and CYP2E1 are proposed to be attractive targets for the development of inhibitors against oxidative stress, a contributing factor in aging, cancer, cardiovascular diseases, diabetes mellitus type 2, and neurodegenerative diseases. Therefore, this study aimed to evaluate the binding abilities of the saponin derivatives on both enzymes eNOS and CYP2E1 using structure-based approaches. An in-house library of 50 saponin derivatives from Vietnamese ginseng was computationally analyzed for their binding affinities and interactions with eNOS and CYP2E1 using Autodock Vina 1.5.6. The results showed that ginsenoside Rc, ginsenoside R3, vina-ginsenoside R20, ginsenoside Re, notoginsenoside R1 and 20(R)-ginsenoside Rh1 established the favorable interactions and exhibited high binding affinities with eNOS and CYP2E1. These compounds are potential candidates for in-vitro and in-vivo assays to assess their promising application in inhibition of these enzymes....

Matrix tablets were developed using moi gum for investigating its suitability for the controlled release using losartan potassium as a model drug. Tablets were prepared by direct compression method. Lactose and dibasic calcium phosphate (DCP) were used as channelling agents. In-vitro studies were performed in 0.1N hydrochloric acid for the first two hours and pH 6.8 phosphate buffer for the next ten hours. The retardation of drug release was influenced by gum concentration and nature of diluents. The drug release was retarded when compared with dissolution patterns of synthetic polymers like polyethylene glycol (PEG 4000 & PEG 6000). The release rate, extent, and mechanisms were found to be governed by the concentration of the gum and channelling agents. Increased rate and extent of the drug release were found by using a higher content of channelling agent in the matrix due to increased porosity. It was found that type and concentration of channelling agent significantly affect the percentage drug release, release rate constant (K) and diffusion exponent (n). The FTIR...

Salbutamol is a bronchodilator, Guaifenesin is an expectorant and Ambroxol is a mucolytic. Combination of these drugs is used in the formulation of cough syrups. In the literature, there is no method reported for the simultaneous estimation of the drugs in oral liquid dosage form. Hence, the present work is aimed to develop reverse phase HPLC method for the simultaneous determination of Salbutamol sulfate (SAL), Guaifenesin (GUA) and Ambroxol hydrochloride (AMB) in oral liquid dosage form and validation of the developed method. The chromatographic separation of the drugs was achieved with the mobile phase system sodium dihydrogen phosphate buffer pH 3.0: acetonitrile: methanol in the ratio of 65:10:25 with the flow rate of 1 mL/min and injection volume 10 µL. An Inertsil C8-3 (250 × 4.6 mm, 5 µm) column was used, and the detection wavelength was 276 nm. This system produced sharp peaks with good resolution, minimum tailing and satisfactory retention times of SAL, GUA and AMB were found to be 3.157, 9.949 and 11.883 min respectively indicating...

A mathematical model optimizing bio-physico-chemical pre-treatment of steam exploded Piper nigrum, and Syzygium aromaticum lignocellulosic biomass waste was developed. The model was developed using, RSM, involving central composite face-centered design (CCD) with six parameters, three levels and 40 runs, at 95% confidence level. Six parameters optimized were quantity of each enzyme (a) Cellulase (ml); (b) Xylanase (ml) for enzymatic actions at 50 ºC; (c) Incubation time of 2-15 days for which substrate were dipped in various solvent systems; (d) Volume of solvent systems used; (e) Time of incubation at given steam pressure for steam explosion; and (f) Steam pressure 10-15 Psi. Cellulase and xylanase enzymes used were with activity (3.563 IU/mL), (33.32 IU/mL) respectively. ANOVA was applied for validation of the predicted model at 95% of confidence level. The Base 10 log transformation was selected from the comparative study, and the model predicted 13.79 µmoles/mL, the release of polyphenols at 2 days of incubation of substrate with 10 ml of the solvent system, under 15.0 psi steam explosion pressure...
