Aim: To evaluate the knowledge, attitude and practice (KAP) related to Adverse Events Following Immunisation (AEFI) of COVID-19 vaccines among MBBS students of Madras Medical College. Materials and Methods: A cross-sectional online survey was conducted using a Google Forms questionnaire circulated via e-mail and WhatsApp. A total of 256 MBBS students participated in the study. Responses related to knowledge, attitude and practice of AEFI reporting were collected and analysed descriptively. Results: Among the respondents, 98% knew the correct expansion of AEFI, 94.1% identified the symptoms of mild AEFI, and 92.7% were aware of cluster AEFI. Regarding attitude, 64.8% agreed that local post-vaccination symptoms resolve spontaneously, and an equal proportion strongly agreed that AEFI reporting contributes to vaccine safety. For practice, 95.7% strongly agreed that they would encourage their peers to report AEFI, and mobile applications were the most commonly used reporting method. Conclusion: MBBS students demonstrated adequate knowledge and a positive attitude towards AEFI associated with COVID-19 vaccines. However, the actual practice of AEFI reporting was relatively lower, indicating...

A vital tool in drug discovery and development, in-silico toxicity prediction tools and algorithms provide economical, time-efficient and morally sound substitutes for traditional in-vitro and in-vivo toxicological testing. Computational techniques are essential to predict the possible toxicity and side effects prior to clinical testing because of the ever-increasing complexity of pharmaceutical compounds and the regulatory bodies’ increasing focus on early toxicity screening. An overview of the in-silico methods for toxicity prediction is discussed in this article, which includes pharmacophore modeling, machine learning algorithms, molecular docking, and Quantitative Structure–Activity Relationship (QSAR) models. It weighs the underlying principles, prediction powersand practical applications of popular database tools including ProTox-II, SwissADME, Toxtree, and ADMETlab. The review also includes case studies that show how the tools are used to evaluate the toxicity of potential drugs. Limitations including model generalizability, lack of standardization, and validation issues persist despite the tool’s increasing dependability. The final section of the analysis looks at the state of regulations today and suggests ways to improve the accuracy and acceptability of...

Background: Fast-dissolving oral films (FDFs) represent a modern drug delivery system designed to disintegrate rapidly in the mouth without water, making them highly suitable for pediatric and geriatric populations. Eucalyptus oil, rich in 1,8-cineole, is utilized for its antimicrobial, anti-inflammatory, and soothing effects on oral and respiratory conditions. Integrating this essential oil into an FDF platform ensures a rapid onset of action and improves patient compliance for managing respiratory relief. Objective: The primary goal of this study was to formulate and characterize eucalyptus oil-loaded FDFs and evaluate the impact of different hydrophilic polymer types and their concentrations on the films’ physicomechanical properties. Methods: Twelve formulations (F1 to F12) were prepared using the solvent casting technique. The study utilized four polymers at varying concentrations: HPMC E5, PVA, NaCMC, and PVP K30. Eucalyptus oil loading (5% w/w), PEG 400 as a plasticizer (15% w/w), and Tween 80 as a surfactant were maintained across formulations. The films were characterized for appearance, thickness, folding endurance, surface pH, moisture content, tensile strength, and disintegration...

The present study aimed to identify potential anti-HIV phytoconstituents from Ocimum tenuiflorum through a novel extraction strategy combined with LC–MS/QTOF-based phytochemical profiling, ADMET and PASS server predictions, HIV target proteins to elucidate bioactive compounds with promising antiviral potential. The LC-MS analysis of the extract revealed the presence of 17 compounds, including flavanones, isoflavones, phenolics, terpenoids, coumarins, and thiazole derivatives. ADMET analysis indicated that most of these phytoconstituents exhibited favourable drug-likeness properties and belonged to toxicity classes IV, V, or VI, suggesting that they are non-toxic and potentially safe for therapeutic use. PASS server predictions revealed that the compounds possess significant HIV integrase-inhibitory...

Due of their ability to damage DNA, genotoxic impurities (GTIs) in active pharmaceutical substances need strict analytical control. Trace process-related GTIs may be present in trazodone hydrochloride, an antidepressant made using multi-step synthetic processes. Nevertheless, there is currently no approved analytical technique for the simultaneous determination of all significant GTIs related to its production. In order to quantify four process-related GTIs in trazodone hydrochloride at sub-ppb levels, a quick and selective UFLC-MS/MS approach was developed and validated. Acetonitrile-5 mM ammonium acetate buffer (pH 5.0, 70:30 v/v) was used for chromatographic separation on an Avantor ACE C18-PFP column (3 µm, 4.6 × 150 mm) under isocratic conditions. Multiple reaction monitoring was used for detection in positive ESI mode. The approach was validated over 0.060-1.200 ppb. For every impurity, linearity was attained with correlation coefficients (R²) ≥ 0.99. Across three levels, accuracy studies produced recoveries ranging from 79 to 102%. Robustness was verified with intentional changes in flow rate and column temperature, and precision satisfied acceptance standards with %RSD < 15%....
