The aim of the present work was to develop and validate a novel, rapid, sensitive, accurate and specific RP-HPLC assay with fluorescence detection for the quantification of amlodipine besylate in rat plasma to be applied to preclinical studies. Flourescein was used as internal standard (IS) and the detection was carried out by an online wavelength switching which enabled detection of amlodipine & flourescein in the same method. The plasma proteins were precipitated by a single step protein precipitation extraction procedure using methanol. Mean recovery of amlodipine besylate from rat plasma was more than 85.0 % for 25 – 5000 ng/ml concentrations. The assay exhibited good linear relationship with an r2 of 0.999. Lower limit of Quantification limit (LLOQ) was 1.84 µg/ml of amlodipine besylate and accuracy and precision were over the concentration range 25 – 5000 ng/ml. Thus, the bionanalytical method for quantification of amlodipine besylate is novel, since use of fluorescence detection & using flourescein as the internal standard has made the method with comparable sensitivity to LC-MS/MS...

The present study was designed to evaluate antidepressant effects of fruit extract of Elaeocarpus ganitrus(75% ethanol)on albino mice. Albino mice of either sex, weighing 25-30g were divided into seven groups and each group contains six animals. The animals were housed in polypropylene cages (38×23×10cm3) with not more than six animals per cage. Group I, II and III were administered orally with distilled water, fluoxetine and imipramine respectively. Group IV, V, VI and VII were administered orally with different doses (20, 40, 60 and 80 mg/kg body weight) of Elaeocarpus ganitrus fruit extract (EGFE). Antidepressant effect of EGFE assessed by forced swim test. The (EGFE) (20 and 40mg/kg) significantly decreased the immobility time and it also increased climbing and swimming time significantly. Duration of immobility of mice increased (246.00±1.73) at high dose (80mg/kg) as compared to vehicle (228.00±4.28). It is concluded that the EGFE exhibit antidepressant effect in tested animal model at low dose but at high dose it is...

A new simple and sensitive reverse phase high performance liquid chromatographic method was developed for simultaneous estimation of Dutasteride and Tamsulosin hydrochloride following forced degradation studies as per ICH guidelines. The degradation products were observed in acid and alkaline hydrolysis for Dutasteride; whereas Tamsulosin hydrochloride showed degradation on exposure to light. All the degradants were separated by preparative thin layer chromatography and subjected to structural elucidation by mass spectrometry using electron spray ionization technique. The base peak of DTS with m/z of 529.1 was formed in positive ESI mode. The daughter ion of DTS was observed at m/z of 461.2. Two degradants with m/z of 546.2 and m/z of 527.5 were formed by acid hydrolysis to give a carboxylic acid derivative and another by dehydrogenation respectively. Degradation product with m/z of 573.3was di-sodium adduct of DTS and was formed only in alkaline hydrolytic condition. The base peak of photo degradant of TMS with m/z of 409.1 was formed in positive ESI mode showed same m/z value of the base...

The aim of the study was to develop and evaluate matrix based controlled drug delivery system of Bupropion hydrochloride. Controlled release tablets were prepared by employing Guar gum, Eudragit RS 100, HPMC K15M, HPMC K100M at different concentration. All 13 batches passed friability, hardness, weight variation, assay but only four batches (CRB7W, CRB8W, CRB10W, and CRB11W) passed the dissolution as per USP 30 NF25 for extended release tablet of Bupropion hydrochloride. Among the four formulation CRB7W & CRB11W follows Higuchi Model and CRB8W & CRB10W follows First order model. The optimized formulations were compared with the marketed product for similarity and dissimilarity factor. CRB7W and CRB10W showed dissimilar result and CRB8W and CRB11W showed similar result with marketed product. HPMC (K15M 7& K100M) as a matrix polymer used in tablet formulation provide a good initial retardation in the release as well as helped to enhance the overall release rate of the drug than that of Gaur gum and Eudragit RS...

Woodfordia fruticosa (L.) Kurz. is a plant with vast medicinal properties. The plant and its parts are reported to be used for the treatment of hemorrhoids, dysentery, diarrhoea, liver diseases, piles, leucorrhoea, menorrhagia, ulcers, wounds, skin diseases, fever, headache, herpes, etc. The present work attempts to evaluate the physicochemical and preliminary phytochemical screening using various extracts of W. fruticosa (Family- Lythraceae). The herbal standardization was carried out on the basis of physico-chemical properties. Physicochemical parameters including ash values, extractive values and fluorescence analysis were evaluated. The qualitative phytochemical screening using various solvent extracts of plant revealed the presence of most of the biologically active phytochemicals in the methanolic extract of Woodfordia fruticosa leaves. TLC profiling of in vivo and in vitro plant part extracts gives an idea about the presence and confirmation of various phytochemicals. TLC screening showed the presence of ellagic acid (Rf 0.54), gallic acid (Rf 0.61), quercetin (Rf 0.8), myricetin (Rf 0.96) and β-sitosterol (Rf 0.58). Results confirmed the presence of therapeutically potent compounds in the...
