Proteomics is the large scale study of proteins. Before going to analyse the differentially expressed proteins, through 2-D gel electrophoresis the proteins were extracted in a sample under stress. In the present study, we were isolated the protein samples of E. coli Nissle 1917 treated with cocoti sap and wine stress. The isolated proteins washed with 2-D clean up kit because it was very sensitive to salts and detergents. Quantify the protein concentration for loading the samples in 2-D electrophoresis. Proteins were separated by based on IEF and Molecular weight. By using Image master 2-D platinum 6.0 software programmes analyze the protein spots in the gel. We noticed over all 800 proteins in our gel, 370 protein spots were visualized clearly, in that considers 15 spots were isolated based on the regulation. Ten spots shows up regulation and remaining shows down regulation. One newly expressed protein was isolated from wine treated gel when compared to the control and also sap treated gels. Independent t-test was performed to analyze the...

Cultures of Aspergillus niger and Bacillus licheniformis were procured from NCL, Pune. Obtained cultures were exposed to UV irradiation. Mutants of Aspergillus niger exposed to 30 and 50 min UV and Bacillus licheniformis exposed to 20 and 30 min UV were selected. Growth of wild type and mutated Aspergillus niger and Bacillus licheniformis was found to be optimum at 48 hrs of incubation time, 30°C of incubation temperature and pH 6. SSF was carried out using two substrates namely banana peel and potato peel. Alpha amylase produced using the two substrates showed good activity but banana peel as a substrate exhibited highest activity of 113.6 (U/ml) in Aspergillus niger mutant exposed to 30 min UV treatment followed by banana peel exhibiting activity of 11.2 (U/ml) in Aspergillus niger mutant exposed to 50 min UV treatment. In this regard, production of amylase which can be used in food & pharmaceutical industry can be increased by using mutants & waste banana...

Liver is a vital organ performing wide range of functions; oxidative damage is implicated in the pathogenesis of various liver disorders. Present study was aimed at evaluating protective ability of Thuja orientalis Linn against thioacetamide induced hepatotoxicity in rats. Thuja orientalis has been used in Ayurvedic medicine for the treatment of various disorders. Thuja orientalis has the biological capabilities of detoxication, antioxidation, and antiinfection. In this study, we evaluated the antihepatotoxic effect ofThuja orientalis aqueous and petroleum ether extract on the thioacetamide induced liver injury in a rat model. Hepatic damage, as reveled by histology and the increased activities of serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) activities, and decreased levels of serum total protein (TP), albumin (Alb) and globulin (G) were induced in rats by an administration of 50 mg/kg thioacetamide was injected intraperitoneally (i.p). Thuja orientalis extract significantly inhibited the elevated AST, ALP and ALT activities and the decreased TP, Alb and G levels caused by thioacetamide intoxication. From this study, it can be...

A rapid and accurate HPLC method was developed for determination of both metformin hydrochloride and pioglitazone hydrochloride in tablet dosage form. The chromatographic separation was conducted on Shimadzu (Prominence LC 20 UFLC XR) connected with PDA detector; using mixed column ODS/Cyano; ACE, (100 x 4.6 mm, 5 µm). The mobile phase was isocratic consisted of Acetonitrile : Phosphate buffer in ratio of (50 : 50 v/v) (buffer was composed of 3.55 gm disodium hydrogen phosphate per liter, adjusted by 85% phosphoric acid to pH 5) and was delivered to the system at a flow rate of 1.2 ml/min. An injection volume of 20 µl was used for pioglitazone hydrochloride and 5 µl for metformin hydrochloride. The detection wavelength (λ max) was 235 nm for metformin HCl and 266 nm for pioglitazone hydrochloride. All assays were performed at ambient conditions. The calibration curve of metformin hydrochloride in mobile phase was linear with correlation coefficient (r2) = 0.99995; over a concentration range of 30 – 750 mg/l for; with a retention...

Complexes of Manganese(II) of general composition Mn(L)2X2 have been synthesized with ligand 2-heptanone semicarbazone and 2-heptanone thiosemicarbazone (where L = 2-heptanone semicarbazone or 2-hepatanone thiosemicarbazone and X=Cl–, SO42-, CH3COO–, NCS–, ClO4–) The IR spectral data of ligands indicate the involvement of sulphur/oxygen and nitrogen in coordination to the central metal. The molar conductance of the complexes of the complexes in fresh solution of DMSO lies in the range of 10 -20 ῼ-1 cm2mol-1 indicating their non-electrolyte behaviour thus the complexes may be formulated as [M(L2)X2].Ligands were characterized by Mass, NMR, and IR. All the complexes were characterized by elemental analysis, magnetic moments, IR, electronic and EPR spectral studies. All the manganese (II) complexes in present study show magnetic moment in the range of 5.90-6.05 B.M; corresponding to five unpaired electrons and hence these are high-spin complexes. The Schiff’s base ligands forms hexacoordinated complexes having octahedral geometry for...
