In this research work, numerous molecules that have substituted triazole phthalazine derivatives were obtained using phthalic anhydride as a starting material. It was taken as starting material that gave reaction in the precence of different conditions with different substituted benzoyl chloride that yielded 6 chloro-3 substituted 6a, 10 a-dihydro-[1, 2, 4] triazolo [3, 4-a] phthalazine derivatives (I-V). These intermediates further reacted with substituted benzaldehyde and yielded 4-2-3-Substituted – 6a, 10 a-dihydro-[1, 2, 4] triazolo [3, 4-a] phthalazin-6-yl) substituted hydrazinyl derivatives (VI-X) which were further react with triethyl amine and acetyl chloride yields (XXI-XXX). Arthritis is a joint disorder which is characterized by inflammation. A joint function to move the body parts connected by its bones. Arthritis means inflammation of one or more joints. The study investigates the anti-arthritic activity in rodents by use of in-vivo parameters Paw volume, Knee diameter, Paw thickness, Paw oedema, and body weight. Some biochemical parameters were also done, such as SGOT, SGPT, Total protein estimation, ESR rate, Haemoglobin level Histopathological investigation. Animals serving as arthritic control received 2% formaldehyde 0.1ml throughout the study; while the animals in the standard group received 10mg/kg of Indomethacin. The animal in the test group received 240mg/kg extract once daily by oral route. To the best of our knowledge, the factorial design methodology has never been used to estimate Thymoquinone (TQ) using the reverse phase High-Performance Liquid Chromatography (RP-HPLC) method, making our work unique. The study aimed to create, verify and validate a simple stability-indicating analytical technique for quantitative detection of TQ in Ayurvedic formulations, black seed and polymeric nanoformulation using Design-experiment® (DOE) version 12. The technique was developed utilizing a Luna C18 HPLC column with a mobile phase of water: methanol (25:75, v/v), administered isocratic ally at 1 ml/min, at 253 nm. The validation parameters were carried out in compliance with ICH criteria using the Quality by Design (QbD) approach. The devised technique was shown to be specific, linear (R2 > 0.999) across the specified concentration range of 0.5 to 16 µg/ml, with detection and quantification limits of 0.02 and 0.08 µg/ml, respectively. Natural products can be the alternative of synthetic medicine and antiviral drugs can be potential from natural products. An attempt was made to evaluate the antiviral activity from the extract of Desmodium gangeticum (L.) DC. as a phytomedicine (GanjhuVirR) for the treatment of dengue fever associated with thrombocytopenia. The present study was evaluated acute and subacute toxicity in rat as well as clinical trial in treated and without treated patients who admitted in hospital due to dengue fever associated with thrombocytopenia. The rats were grouped into group 1 (control) and 2, 3 and 4 (treated) for toxicity test while 51 patients were included in the clinical trial and 25 were treated and 26 provided only standard management as control. The parameters viz. platelet count, fever profile, average hospitalization period, clinical profiles and viral load reduction were tested for both the group. Nutraceutical is employed to explain a medicinal or nutritional component with a food, plant, or natural material that will be purified or concentrated. It is used to improve health by preventing or treating disease. Non-alcoholic fatty liver disease (NAFLD) is a clinical condition characterized by lipid infiltration of the liver with a high prevalence in the general population affecting 25% of adults with a double prevalence in diabetic and obese patients. Nearly 1/3 of NAFLD progresses to non-alcoholic steatohepatitis (NASH), leading to liver fibrosis and cirrhosis. There are currently no specific drugs approved on the market to treat NAFLD. So the treatment is essentially based on lifestyle optimization.
