Pseudomonas aeruginosa is a Gram-negative pathogenic bacterium that can cause a variety of hospital-acquired diseases. It is a multidrug-resistant bacterium and can survive a wide range of antibiotics; hence there is a concern about its limited therapy options. In this scenario, the discovery and development of novel antibiotics are in high demand. In our research work, molecular docking a Bioinformatics technique, is applied to find the ideal lead molecule from plants. For this study, we have extracted literature-proven phytocompounds from a variety of traditional medicinal plants and their structures were retrieved from the PubChem database. Lipinski and ADME/TOX screening were used to evaluate drug likeliness, followed by docking against the target protein. Pseudomonas aeruginosa elastase, or LasB is a virulence protease that hydrolyzes host tissue and is thus considered the target protein. Among different neoplastic lesions, hepatocellular carcinoma plays a pivotal role due to the extreme difficulty of treating it in advanced conditions. Research shows that there is a high intractability towards chemotherapy for this disease. Therefore, using medicinal plants can be another approach to fend off this grim scenario and pave a new way to prolong survival time. For this purpose, the ethanolic extracts of leaves and fruits of Cassia fistula (Golden shower) as a probable anticancer drug have been administered in the liver cancer cell line, HEPG2 and the gene expression of cytokines (interleukin-6, interleukin-8, interleukin IL-1β, transforming growth factor-TGF-β1, tumour necrosis factor-TNF-α) against this treatment has been analyzed in the present work. Gouty arthritis, an inflammatory joint disease, occurs due to the accumulation of Monosodium Urate (MSU) crystals in the joints, mainly at the metatarsophalangeal joints of the big toe. At present, the incidence and prevalence of gout are increasing globally. The various risk factors are responsible for the development of gouty arthritis, including nonmodifiable, modifiable, and genetic factors. Under physiological conditions, the purine undergoes a catabolic pathway to produce the uric acid end product excreted from the body using various uric acid transporters. The overproduction or under-excretion of uric acid leads to the pathophysiology of hyperuricemia and promotes the MSU crystals formation and deposition in joints to trigger inflammation by releasing inflammatory mediators in gouty arthritis. Objective: The study aims to evaluate the granuloma and associated inflammatory mediators inhibition potential of the chloroform extract of Piper pedicellatum C. DC. Leaves, as plant leaves, are being used traditionally in the mitigation of various inflammatory conditions; however, there is an insufficiency of scientific evidence about the relevant bioactivity of the plant. Method: The cotton pellet induced-granuloma model in rats is used in the study, providing insights into subsequent phases of inflammation. Piper pedicellatum C. DC. Leaf chloroform extract at 200 mg/kg and 400 mg/kg and indomethacin (reference standard, 10 mg/kg) were used during treatment. Results: The chloroform extract at 200 mg/kg and 400 mg/kg dose inhibited exudate formation by 25.23% and 38.43%, respectively, and inhibition of granuloma tissue was found to be 27.62% and 40.33%, respectively. Further significant decrease in serum inflammatory markers CRP and PGE2 and reduced cytokine TNF-α and IL-6 were observed in chloroform extract and indomethacin-treated rats.
